柴芍乳癖颗粒对乳腺非典型增生大鼠MVD及VEGF蛋白表达的影响

摘  要:目的:研究柴芍乳癖颗粒对大鼠乳腺非典型增生病变血管生成及相关因子的干预作用,探讨柴芍乳癖颗粒防治乳腺非典型增生病的机理。方法:将60只SD雌性大鼠随机分为空白对照组(10只)及模型制备组(50只)。采用7,12-二甲基苯并蒽(7,12-dimethyltetraphene,DMBA)灌胃诱导乳腺非典型增生病变模型,病理学验证造模成功后,将45只造模成功大鼠随机分为模型组、阳性对照组、中药低剂量组、中药中剂量组、中药高剂量组,每组9只。药物干预30 d,干预后第2天处死所有大鼠,切取第二、三对乳腺组织,采用SP免疫组化法检测病变组织微血管密度(micro vessel density,MVD)及血管内皮生长因子(vascular endothelial growth factor,VEGF)的表达情况并行病理学检查。结果:(1)病理学:与模型组比较,中药高剂量组及阳性对照组大鼠乳腺组织病理形态学分布较前好转(P<0.05),癌前病变大鼠数量下降;(2)免疫组化:与模型组比较,中药中、高剂量组及阳性对照组大鼠乳腺组织MVD表达降低(P<0.05),阳性对照组及中药高剂量组大鼠VEGF阳性率下降(P<0.05);阳性对照组及中药高剂量组比较,差异无统计学意义(P>0.05)。结论:柴芍乳癖颗粒及他莫昔芬可改善乳腺非典型增生大鼠乳腺组织病理形态学表现,可显著抑制乳腺非典型增生模型大鼠VEGF表达并可降低MVD,可有效防治乳腺癌前病变,且柴芍乳癖颗粒以中、高剂量疗效更好。Objective:To study the intervention effect of Chaishao Rupi granules on angiogenesis and related factors of mammary atypical hyperplasia in rats,and to explore the mechanism of preventing and treating mammary atypical hyperplasia by Chaishao Rupi granules.Methods:Totally 60 SD female rats were divided into blank group(n=10)and model preparation group(n=50).The rats in model preparation group were given 7,12-dimethyltetraphene(DMBA)intragastric induction to induce breast atypical hyperplasia lesion model.After successful pathological verification of modeling,45 successfully modeled rats were randomly divided into model group,positive group,low dose group of Chinese medicine(low dose group),medium dose group of Chinese medicine(medium dose group),and high dose group of Chinese medicine(high dose group),with 9 rats in each group.After 30 days of drug intervention,all rats were euthanized on the second day after intervention.The second and third pairs of mammary tissues were cut.The expression of micro vessel density(MVD)and vascular endothelial growth factor(VEGF)was detected by SP immunohistochemistry and pathological examination was performed.Results:According to pathomorphology,the pathological and morphological distribution of rat mammary gland tissue has improved in high dose group and positive group compared to model group(P<0.05),and the number of rats with precancerous lesions has decreased.According to immunohistochemistry,the expression of MVD in rat breast tissue decreased in high dose group,medium dose group and positive group compared to model group(P<0.05),and the positive expression intensity of VEGF decreased in positive group and high dose group(P<0.05).There was no statistically significant difference between the positive group and high dose group(P>0.05).Conclusion:Chaishao Rupi granules and Tamoxifen can improve the histopathologic features of breast in rats with atypical hyperplasia of breast,significantly inhibit the expression of VEGF and reduce MVD.Chaishao Rupi granules and Tamoxifen can eff
