甘草种子的基源鉴定及质量评价研究

摘  要:目的:鉴别甘草种子的真伪及其基源,评价分子条形码对甘草种子的鉴定可行性,并对甘草种子进行质量评价,确定野生甘草种子和栽培甘草种子的质量差异。方法:利用ITS和psbA-trnH条形码,对13批甘草种子进行真伪和基源鉴定,并通过测定甘草种子的长度、宽度、厚度、千粒重、净度及发芽率,评价5批野生甘草种子和5批栽培甘草种子的质量。结果:利用ITS和叶绿体条形码psbA-trnH相结合的方法可以对甘草种子的真伪及基源进行鉴定,在13批甘草种子中有2批为光果甘草,11批为乌拉尔甘草;野生甘草种子的长度、宽度、厚度、千粒重、发芽率、净度及含水量高于栽培甘草种子,差异均有统计学意义(P<0.05或P<0.01)。结论:利用ITS和psbA-trnH条形码可准确鉴别甘草种子的基源,且野生甘草种子在播种品质方面均优于栽培甘草种子。Objective:To identify the authenticity and origin of licorice seeds,and evaluate the feasibility of molecular barcode identification of licorice seed.To evaluate the quality of wild licorice seeds and cultivated licorice seeds to determine the quality difference.Methods:Using ITS and psbA-trnH barcodes,13 batches of licorice seeds were identified for authenticity and origin.The quality of 5 batches of wild and 5 batches of cultivated licorice seeds was evaluated by measuring the length,width,thickness,1000 kernel weight,purity and germination rate of the seeds.Results:The ITS sequence and psbA-trnH sequence can be used to identify the authenticity and origin of glycyrrhiza seeds.Among 13 batches of glycyrrhiza seeds,2 batches were Glycyrrhizaglabra and 11 batches were Glycyrrhizauralensis.There were significant differences in length,width,thickness,1000 grain weight,water content,germination rate and purity between wild licorice seeds and cultivated licorice seeds(P<0.05 or P<0.01).Conclusion:The use of ITS and psbA-trnH barcode can accurately identify the source of glycyrrhiza seeds,and the wild glycyrrhiza seeds are superior to the cultivated glycyrrhiza seeds in terms of sowing quality.
