摘 要:棉蚜Aphis gossypii Glover种群出现明显分化,表现在生物型间的寄主偏好差异及单倍型间基因组层面分化。为了解棉蚜种群分化过程中激素合成相关基因的演变,选择Hap1和Hap3型棉蚜的法尼醇脱氢酶(HGO)进行分析。根据本实验室发布的两种单倍型棉蚜的基因组信息,在Hap1和Hap3型棉蚜基因组中各鉴定出5个AgosHGO基因,其中4个为重复拷贝,位于常染色体A1,另外1个位于常染色体A2。AgosHGO基因的CDS序列、基因结构在不同单倍型棉蚜间存在变异。在Hap3型棉蚜中,位于A1染色体上AgosHGO基因分布的区域存在一个Gypsy转座子的插入。利用NCBI公共数据已发布的棉蚜SRA数据,分析得到AgosHGO表达量与棉蚜的取食寄主显著相关,但不随体色及温度发生改变;AgosHGO3表达量在啶虫脒处理后显著下调50%;雄蚜成蚜期AgosHGO5表达量较若蚜期有显著上调,达14~20倍。本研究证实HGO基因在Hap1型和Hap3型棉蚜间出现分化,且其表达量能响应自身发育和外界环境变化,研究结果对于揭示棉蚜种群分化过程中激素响应的调控机制,开发基于激素调控的棉蚜生物防治技术具有重要的意义。Cotton aphid,Aphis gossypii Glover,differentiated into biotypes,showing obvious differences in host preference among the biotypes and differentiation at the genome level.In order to understand the evolution of hormone synthesis-related genes during the biotype differentiation of cotton aphids,farnesol dehydrogenase(HGO)of Hap1 and Hap3 cotton aphids was selected for analysis.According to the genome information of the two biotypes released by our laboratory,five AgosHGO genes were identified in each of the genomes of Hap1 and Hap3 cotton aphids,four of which are repetitive copies locate on chromosome A1 and the other one locates on chromosome A2.The position of the HGO gene on the chromosome,gene coding and sequence composition were analyzed to obtain relevant evolution information.The SRA data of cotton aphid published by NCBI were used to analyze the expression changes of HGO gene under different treatment conditions.There were differences in CDS sequence and gene structure of AgosHGO among different biotypes.In the Hap3 cotton aphid,a Gypsy transposon insertion is in the region of AgosHGO gene which distributes on chromosome A1.The analysis showed that the expression of AgosHGO was significantly different among different hosts but did not change with body color and temperature.The expression of AgosHGO3 was significantly down-regulated by 50%after acetamiprid treatment.The expression of AgosHGO5 in male aphid was significantly up-regulated in adult instar compared with the 1st—4th instar,up to 14—20 times.This study confirms that the HGO gene differentiates in Hap1 and Hap3 cotton aphids,its expression level can respond to its own development and changes in the external environment.It is of great significance to develop biological control technologies based on the regulation of hormone.
