摘  要:目的探究四君子汤(SJZD)对丝裂霉素C(mitomycin C,MMC)诱导的免疫毒性的减毒作用.方法体外分离培养小鼠脾淋巴细胞,并采用刀豆蛋白A(ConA)刺激细胞增殖,选用系列浓度的MMC、SJZD以及两药联用分别与脾淋巴细胞共培养,采用MTT法检测不同药物对细胞活力的影响,采用ELISA法检测不同药物对脾淋巴细胞培养基上清液的细胞因子白细胞介素-2(IL-2)和干扰素-γ(IFN-γ)水平的影响.结果培养24和48 h后,MMC组与对照组(CON)相比细胞活力均显著下降,且48 h下降更明显(∗P<0.05,∗∗P<0.01);培养24 h后,SJZD组在25~200μg·mL^(-1)内与CON组相比可促进细胞生长,100μg·mL^(-1)时细胞活力显著上升(∗P<0.05),在质量浓度400~800μg·mL^(-1)内抑制细胞生长,SJZD(25~800μg·mL^(-1))联用MMC组与MMC组相比细胞活力上升;培养48 h后,SJZD组在25~200μg·mL^(-1)内与CON组相比可促进细胞生长,100~200μg·mL^(-1)时细胞活力显著上升(∗P<0.05),在质量浓度400~800μg·mL^(-1)内抑制细胞生长,COM组与MMC组相比细胞活力上升,其中100~200μg·mL^(-1)内显著上升(#P<0.05);MMC组IL-2和IFN-γ水平与CON组相比显著下降,SJZD(100、200μg·mL^(-1))组与CON组相比显著上升(∗P<0.05,∗∗P<0.01),COM组与MMC组相比IL-2和IFN-γ水平显著上升(#P<0.05,##P<0.01).结论SJZD对免疫细胞的生长及细胞因子(IL-2和IFN-γ)的分泌具有很好的促进作用,可以在一定程度减弱MMC对脾淋巴细胞的毒性以及对细胞分泌IL-2和IFN-γ的抑制作用.Objective To explore the attenuation effect of Sijunzi Decoction(SJZD)on immunotoxicity induced by mitomycin C(MMC).Methods Mouse spleen lymphocytes were isolated and cultured in vitro.Conanin A(ConA)was used to stimulate cell proliferation.MMC,SJZD and the co-treatment were used to co-culture with spleen lymphocytes respectively.ELISA was used to detect the effects of different drugs on the levels of interleukin-2(IL-2)and interferon-γ(IFN-γ)in splenic lymphocyte culture medium supernatant.Results Compared with control group(CON),the cell viability of MMC group was significantly decreased after 24 and 48 h,and the decrease was more obvious at 48 h(∗P<0.05,∗∗P<0.01).After 24 h,SJZD group promoted cell growth in the range of 25-200μg·mL^(-1) and cell viability was prominently increased at 100μg·mL^(-1)(∗P<0.05),inhibited cell growth in the range of 400-800μg·mL^(-1),and SJZD(25-800μg·mL^(-1))combined with MMC group(COM)increased cell viability compared with MMC group.After 48 h,SJZD group promoted cell growth in the range of 25-200μg·mL^(-1) and cell viability was significantly increased at 100-200μg/mL(∗P<0.05),inhibited cell growth in the range of 400-800μg·mL^(-1).The cell viability of COM group(100-200μg·mL^(-1))was obviously increased compared with that of MMC group(#P<0.05).Compared with CON group,the levels of IL-2 and IFN-γin MMC group were prominently reduced,SJZD(100 and 200μg·mL^(-1))groups were significantly increased(∗P<0.05,∗∗P<0.01).COM groups were significantly increased compared with MMC groups(#P<0.05,##P<0.01).Conclusion SJZD can promote the growth of immune cells and the secretion of cytokines(IL-2 and IFN-γ),reduce the inhibition of MMC to splenic lymphocytes and the secretion of IL-2 and IFN-γto a certain extent.
