摘 要:多杀菌素是刺糖多孢菌(Saccharopolyspora spinosa)经有氧发酵产生的一种大环内酯类抗生素。为提升刺糖多孢菌生产多杀菌素能力,本文通过对刺糖多孢菌工程菌SAS-004进行常压室温等离子结合抗性前体甘氨酸、紫外结合抗性前体丙酸钠和亚硝基胍结合抗性前体2-脱氧-D-葡萄糖三种方式选育,成功筛选到一株遗传稳定性良好的高产突变菌株809-5E,其多杀菌素产量相较于原始工程菌SAS-004提升1.14倍,与野生型菌株ATCC49460相比增加10倍,为工业化生产多杀菌素的菌株选育提供了重要参考。Spinosad is an original macrolide antibiotic from the aerobic fermentation of Saccharopolyspora spinosa.In this study,the engineering strain SAS-004 of Saccharopolyspora spinosa was systematically selected using three methods in room temperature plasma combined with the resistant precursor glycine,UV radiation combined with the resistant precursor sodium propionate,and N-methyl-N’-nitro-N-nitrosoguanidine combined with the resistant precursor 2-deoxy-D-glucose,which were employed to enhance the strain’s resistance and productivity.A high-yield and favorable genetic stability mutant strain of Saccharopolyspora spinosa was successfully identified,in which the spinosad production increased 1.14-fold compared to the original engineered strain SAS-004 and 10-fold compared to the wild type strains ATCC49460,respectively.These findings provide some valuable references for the strain selection in industrial production of spinosad.
