摘  要:采用柠檬酸钠-硫酸铜方法制备若羌红枣多糖铜螯合物(RJP-Cu),并对其含量、表征、安全性、补铜效果及免疫增强活性进行综合检测分析,其中安全性试验选择36只SPF级昆明(KM)小鼠,随机分为空白对照组(生理盐水)和灌胃2300、1150、575、288、144 mg/kg RJP-Cu试验组。结果显示:RJP-Cu与若羌红枣多糖(RJP)相比,多糖含量、蛋白含量未发生明显变化,糖醛酸含量下降,铜含量从2 mg/kg提升至583 mg/kg,RJP-Cu电镜结构发生明显变化,RJP-Cu同时具有多糖和铜螯合吸收峰;安全性评价结果显示,RJP-Cu的安全使用浓度为575 mg/kg,该浓度下小鼠血常规各项指标与空白对照组无显著性差异(P>0.05),肝脏、肾脏和肠道无病理变化。RJP-Cu补铜效果呈浓度依赖性,随着RJP-Cu浓度提高,血清、肝脏和肌肉中铜浓度提升;RJP-Cu低剂量(144 mg/kg)组相对空白对照组能够显著促进小鼠免疫球蛋白(Ig)A、IgG、白细胞介素(IL)-2、IL-6、肿瘤坏死因子α(TNF-α)的分泌(P<0.05),表现出良好的免疫增强活性。结论:RJP-Cu螯合成功,144 mg/kg低剂量的RJP-Cu具有良好的免疫增强作用。his study was to prepare copper chelates of polysaccharides from Ruoqiang red jujube(RJP-Cu)and detect their biological activity.RJP-Cu was prepared using the sodium citrate copper sulfate method,and comprehensive analysis was conducted on various contents,characterization,safety,copper supplementation effect,and immune enhancement activity of the copper chelates.In the safety test,36 SPF KM mice were selected and randomly divided into blank control group(normal saline)and 2300,1150,575,288,144 mg/kg RJP-Cu experimental groups.The results showed that,compared with RJP,the polysaccharide content and protein content in RJP-Cu did not change,the uronic acid content in it decreased,the Cu content increased from 2 mg/kg to 583 mg/kg,the electron microscope structure of RJP-Cu changed significantly,and RJP-Cu had both polysaccharide and Cu chelating absorption peaks.The safety test results showed that the safe concentration of RJP-Cu was 575 mg/kg.At this concentration,there was no significant difference in blood routine indexes compared with the blank control group of mice(P>0.05),and no pathological changes in the liver,kidney and intestine of the rodents.The effect of RJP-Cu supplementation was concentration-dependent.With the increase in RJP-Cu concentration,the Cu concentration in the serum,liver and muscle of the animals increased.The low-dose(144 mg/kg)RJP-Cu group significantly promoted the secretion of IgA,IgG,IL-2,IL-6 and TNF-αin the tested mice,compared with the blank control group(P<0.05),which exhibited good immune-enhancing activities.These results indicated that RJP-Cu chelated successfully,and the low dose(144 mg/kg)of RJP-Cu had a good immune enhancement effect.
