摘 要:目的:通过网络药理学方法探索涤痰汤在脑卒中治疗中的分子机制,为其在未来脑卒中治疗中的应用和进一步的实验室研究提供科学依据和参考。方法:利用TCMSP数据库,筛选涤痰汤中的活性成分,并预测其潜在靶点。使用SwissTargetPrediction数据库和UniProt数据库对这些靶点进行验证和修正。通过Cytoscape软件构建“中药—成分—靶点”网络图,进而使用R语言中的clusterProfiler包进行GO富集和KEGG通路分析,以揭示涤痰汤的作用通路。结果:研究发现涤痰汤共有2 995个活性物质和447个潜在作用靶点。这些活性成分主要关联了调控凋亡、血管功能改善、抗炎作用等生物学过程。涤痰汤的关键作用靶点包括GAPDH、STAT3、TNF等,这些蛋白质在脑卒中的病理过程中发挥重要作用。KEGG通路分析揭示了涤痰汤可能影响的主要信号通路,如癌症信号通路、脂质与动脉粥样硬化等。结论:涤痰汤通过多种生物学通路和靶点发挥治疗脑卒中的作用,显示出显著的抗炎、抗氧化和血管功能改善等效果。这些结果为涤痰汤的临床应用提供了分子机制上的证据,支持了其在传统中医治疗脑卒中中的有效性。Objective:To explore the molecular mechanisms of ditan decoction in the treatment of stroke through network pharmacology methods,providing scientific evidence and references for its application in future stroke treatment and further laboratory research.Methods:Using the TCMSP database,active components within ditan decoction were screened and their potential targets predicted.These targets were verified and refined using the SwissTargetPrediction and UniProt databases.A“Traditional Chinese Medicine-Component-Target”network diagram was constructed using Cytoscape software,followed by GO enrichment and KEGG pathway analyses using the clusterProfiler package in R,to reveal the action pathways of ditan decoction.Results:The study identified 2995 active substances and 447 potential targets in ditan decoction.These active components are mainly associated with biological processes such as apoptosis regulation,vascular function improvement,and anti-inflammatory actions.Key targets of ditan decoction include GAPDH,STAT3,and TNF,which play significant roles in the pathological processes of stroke.KEGG pathway analysis revealed major signaling pathways that ditan decoction may affect,such as cancer signaling pathways and lipid and atherosclerosis.Conclusion:Ditan decoction exerts effects on stroke treatment through various biological pathways and targets,demonstrating significant anti-inflammatory,antioxidative,and vascular function improvement effects.These results provide molecular evidence for the clinical application of ditan decoction,supporting its effectiveness in traditional Chinese medicine treatment of stroke.
