摘 要:目的探究Sp3的SUMO化修饰在细粒棘球蚴感染的巨噬细胞中的作用。方法体内随机分为对照组(PBS)与实验组(肝被膜下接种原头节悬液),免疫组化检测小鼠肝脏SUMO1、UBC9表达;免疫荧光双染检测巨噬细胞CD206和SUMO1荧光共定位。体外RAW264.7巨噬细胞分为对照组、LPS组、LPS+EgCF组,12 h收集细胞qRTPCR检测SUMO1、UBC9、IL-6、IL-10 mRNA表达;免疫印迹法检测SUMO1、UBC9的蛋白水平;COIP检测SUMO1与Sp3的互作。结果体内实验:与对照组比,实验组包囊近端肝脏组织炎性区细胞高表达SUMO1、UBC9蛋白,CD206+巨噬细胞与SUMO1共定位。体外实验:EgCF刺激RAW264.7细胞炎症模型促进IL-10表达,抑制IL-6表达,诱导SUMO1、UBC9 mRNA、蛋白表达,诱导Sp3的SUMO化。结论细粒棘球蚴感染诱导巨噬细胞Sp3的SUMO化修饰。Objective To investigate the role of SUMO modification of SP3 in macrophages infected with Echinococcus granulosus.Methods In vivo,the mice were randomly divided into control group(PBS)and experimental group(subcapsular suspension of the original head segment was inoculated).The expressions of SUMO1 and UBC9 in liver were detected by immunohistochemistry.Fluorescence co-localization of macrophage CD206 and SUMO1 was detected by immunofluorescence double staining.RAW264.7 macrophages were divided into control group,LPS group and LPS+EgCF group.qRTPCR was collected at 12 h to detect the mRNA expression of SUMO1,UBC9,IL-6 and IL-10.The protein levels of SUMO1 and UBC9 were detected by Western blot.COIP detects the interaction between SUMO1 and Sp3.Results In vivo experiment:Compared with the control group,SUMO1 and UBC9 proteins were highly expressed in the inflammatory area of liver tissue near the cyst in experimental group,and CD206+macrophages were co-localized with SUMO1.In vitro experiments:EgCF stimulated the inflammatory model of RAW264.7 cells to promote IL-10 expression,inhibit IL-6 expression,induce SUMO1,UBC9 mRNA and protein expression,and induce SUMO of Sp3.Conclusion Echinococcus granulosus infection induces SUMO modification of macrophage Sp3.
