摘  要:细胞的机械性能与其生理状态和功能密切相关,本文基于库尔特原理研制了细胞检测系统。以斑马鱼胚胎细胞为检测对象,设计制作了带有收缩结构的微流控通道,利用检测系统测量细胞通过微收缩通道时通道两端的阻抗变化,通过记录信号幅值变化与通过时间来表征细胞电学特性与力学特性。实验结果证明:在低频信号下,该系统测得其通过收缩通道引起的幅值与时间变化主要取决于细胞体积大小,验证了健康斑马鱼胚胎细胞具有绝缘特性,并根据斑马鱼胚胎细胞膜的选择透过性与细胞活性的关系,实现了对斑马鱼胚胎细胞的活性检测;在高频信号下,对斑马鱼胚胎细胞进行跟踪检测,结果表明3个不同生长阶段下测得电压幅值变化具有明显差异。The mechanical properties of cells are closely related to their physiological state and function.A cell detection system is researched and fabricated based on the Coulter principle.Zebrafish embryo cells are used as the detection object,and microfluidic channels with contraction structures are designed and fabricated.The detection system is used to measure the impedance changes at both ends of the channel when cells passing through the micro⁃contraction channel.By recording signal amplitude change and passing time,characterize the electrical and mechanical properties of the cells.The experimental results show that under low⁃frequency signals,the amplitude and time changes caused by passing through the contraction channel mainly depend on the cell volume size,verifying the insulating characteristics of healthy zebrafish embryo cells.According to the relationship between the selective permeability of the zebrafish embryo cell membrane and cell activity,the activity detection of zebrafish embryo cells is achieved.Under high⁃frequency signals,zebrafish embryo cells are tracked and measured and the result shows that the measured the voltage amplitude changes have significant differences among three different growth stages.
