摘 要:目的建立鉴定强骨饮化学成分的超高效液相色谱-四极杆-飞行时间串联质谱(UPLC-Q-TOF-MS/MS)法。方法色谱柱为Kromasil 100-5-C_(18)柱(250 mm×4.6 mm,5μm),流动相为0.1%甲酸水溶液-乙腈(正离子模式)和0.5 mmol/L乙酸铵水溶液-乙腈(负离子模式),梯度洗脱,在正、负离子模式下对强骨饮进行质谱检测。根据质谱仪捕捉的化合物精准相对分子质量及二级碎片离子信息,与Agilent TCM Library-V20-04-17数据库的信息比对及查阅相关文献,对强骨饮的化学成分进行定性分析。结果共鉴定出35种化学成分,其中有机酸类10种,黄酮类、环烯醚萜类各4种,酯类3种,糖苷类、皂苷类、氨基酸类、生物碱类、核苷类、色酮类各2种,黄烷醇类、嘌呤类各1种。结论该研究首次对强骨饮的化学成分进行了全面定性分析,为后期质量控制和确定其可能的药效物质基础研究提供了参考。Objective To establish an ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS/MS) method for identifying the chemical components of Qiangguyin.Methods The chromatographic column was Kromasil 100-5-C_(18)column(250 mm × 4.6 mm,5 μm),the mobile phase was 0.1% formic acid aqueous solution-acetonitrile(positive ion mode) and 0.5 mmol/L ammonium acetate aqueous solution-acetonitrile(negative ion mode) with gradient elution.Mass spectrometry detection of Qiangguyin was performed under the positive and negative ion modes.Based on the precise relative molecular weight and secondary fragment ion information of the components captured by the mass spectrometer,combined with the Agilent TCM Library-V20-04-17 database,the chemical components of Qiangguyin were qualitatively analyzed by comparing and consulting relevant literature.Results A total of 35 chemical components were identified,including ten organic acids,four flavonoids and four iridoid,three esters,two glycosides,two saponins,two amino acids,two alkaloids,two nucleosides,two chromones,one flavanol,and one purine.Conclusion This study conducted a comprehensive qualitative analysis of the chemical components of Qiangguyin for the first time,which provides a reference for the quality control and determination of the possible pharmacological substance basis research of this product.
