摘  要:目的为使用威灵仙治疗风湿性关节炎提供参考。方法通过TCMSP,Drugbank,SwissTargetPrediction,SEA数据库预测药物活性成分和靶点,通过GeneCards和OMIM数据库筛选风湿性关节炎靶点,通过Venny 2.1.0在线作图平台获得共有靶点,通过String数据库构建蛋白-蛋白互作(PPI)网络,通过Cytoscape 3.9.1软件对PPI网络进行拓扑学分析,通过Metascape数据库进行基因本体(GO)功能分析和京都基因及基因组百科全书(KEGG)通路富集分析。结果筛选出8种威灵仙活性成分,主要为3-表齐墩果酸、β-谷甾醇、豆甾醇、三萜皂苷A等,涉及潜在靶点362个。筛选出风湿性关节炎潜在靶点1164个,获得药物活性成分-疾病共有靶点112个。PPI网络分析结果显示,核心靶点为白细胞介素6、肿瘤坏死因子、白细胞介素1B、前列腺素过氧化物酶2、基质金属蛋白酶9。GO功能分析结果显示,共获得条目85条,包括生物过程40条、细胞组分19条、分子功能26条,主要通过内肽酶活性、氧化还原酶活性等分子功能发挥作用。KEGG通路富集分析结果显示,威灵仙对花生四烯酸代谢通路、磷脂酰肌醇-3-激酶-丝氨酸/苏氨酸蛋白激酶信号通路、细胞程序性死亡通路等产生影响。结论威灵仙通过多成分、多靶点、多通路调节机体免疫和炎性反应,改善关节功能,抑制程序性死亡细胞发展等,从而达到治疗风湿性关节炎的作用。Objective To provide a reference for the application of Clematidis Radix et Rhizoma in the treatment of rheumatoid arthritis.Methods Active ingredients and targets were predicted by the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),Drugbank,SwissTargetPrediction and SEA database.GeneCards and OMIM were used to screen the targets of rheumatic arthritis.The common targets were obtained through the Venny 2.1.0 online mapping platform.The protein-protein interaction(PPI)network was constructed by the String database.Topological analysis of PPI was carried out by the Cytoscape 3.9.1 software.Gene ontology(GO)functional analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed by the Metascape database.Results Eight active ingredients of Clematidis Radix et Rhizoma were screened,mainly including 3-epioleanolic acid,β-sitosterol,stigmasterol,triterpenoid saponin A,involving 362 potential targets.A total of 1164 potential targets of rheumatoid arthritis were screened,and 112 active ingredient-disease targets were obtained from medicinal herbs.The PPI network analysis results showed that the core targets were interleukin-6,tumor necrosis factor,interleukin-1B,prostagllandin-endoperoxide synthase 2(PTGS2),and matrix metalloproteinase protein 9(MMP9).The GO functional analysis results showed that 85 items were obtained,including 40 biological processes,19 cellular components,and 26 molecular functions,mainly through molecular functions such as endopeptidase activity and oxidoreductase activity.The KEGG pathway enrichment analysis results showed that Clematidis Radix et Rhizoma had an effect on the arachidonic acid metabolism pathway,phosphatidylinositol-3-kinase-serine/threonine protein kinase(PI3K-Akt)signaling pathway,and programmed cell death pathway.Conclusion Clematidis Radix et Rhizoma regulates the immune and inflammatory responses of the body through multiple components,targets,and pathways to improve joint function,inhibit the dev
