摘 要:基于纳米微孔的库尔特测量方法可以将粒径测量下探到100 nm甚至更小,文章研究分别用标准聚苯乙烯微球和虾青素评估基于纳米微孔的库尔特法粒径测量结果,数据表明对标准微球粒径测量误差优于±10%,重复性优于1%;对虾青素的测量,应关注样品的稀释溶液盐离子浓度、前处理振荡频率变化可能导致样品破乳絮凝形成大颗粒,导致测量粒径偏大,样品粒径分布也发生偏移。在使用基于纳米微孔的库尔特分析仪测量生物制品时,应关注稀释液盐离子浓度、样品前处理条件对测量误差的影响,通过优化方法及处理工艺,保证测量结果准确。The Coulter measurement method based on nanopores can measure particle size down to 100 nm or even smaller.In this study,the standard polystyrene microspheres and astaxanthin are used to evaluate the particle size measurement results of the Coulter method based on nanopores.As a result,the data shows that the measurement error of standard microsphere particle size is better than±10%,and the repeatability is better than 1%;for the measurement of astaxanthin,attention should be paid to the salt ion concentration of the sample's dilute solution and oscillation frequency changes of the pretreatment,which may cause sample damage and form large particles.These may result in the larger sample size and size distribution shifting.It should pay attention to the impact of the salt ion concentration of the diluent and sample pretreatment conditions on the measurement error when using a nanopore⁃based Coulter analyzer to measure biological products.The accuracy of the measurement results can be ensured by optimizing methods and processing techniques.
