基于网络药理学探讨柴胡-黄芩“异病同治”功能性消化不良和抑郁症的作用机制

摘  要:目的运用网络药理学方法探讨柴胡-黄芩“异病同治”功能性消化不良和抑郁症的作用机制。方法通过TCMSP数据库和SuperPred数据库获取柴胡-黄芩的活性化合物和作用靶点,通过GeneCards数据库和OMIM数据库获取功能性消化不良和抑郁症的靶点,同时得到柴胡-黄芩异病同治的共同靶点。通过Cytoscape 3.9.1软件和String数据库构建“药对-活性成分-共同靶点”网络、蛋白互作网络,找出关键靶点和核心成分;对共有靶点进行GO和KEGG通路富集分析,运用Autodock软件进行分子对接验证。结果筛选出柴胡-黄芩53个化学成分,其治疗两种疾病共有靶点93个,获得2091条GO功能条目和135条KEGG通路,包括HIF-1、内分泌抵抗等通路;最后,分子对接结果表明关键活性成分与核心靶点之间具有很强的亲和力。结论柴胡-黄芩可能是通过调节炎症反应、改善胃肠功能发挥“异病同治”作用。OBJECTIVE To explore the mechanism of Bupleurum-Scutellaria in treating functional dyspepsia and depression with the method of network pharmac ology.METHODS The active compounds and targets of Bupleurum-S cutellaria were obtained through TCMSP and SuperPred.The targets of functional dyspepsia and depression were obtained from Genecards Database and OMIM database,and the targets of Bupleurum-Scutellaria for treating depression and function al dyspepsia were also obtained.Cytoscape 3.9.1 software and String database were used to constructe“drug pair-active component-common target”and PPI ne twork to find out the key targets and core components;the common targets were a nalyzed by KEGG pathway enrichment analysis and GO enrichment analysis,and Auto Dock software was used for molecular docking.RESULTS A total o f 53 active components of Bupleurum-Scutellaria and 93 common targets in the tr eatment of functional dyspepsia and depression were collected.2091 GO functiona l items and 135 KEGG pathways were obtained by KEGG pathway analysis and GO enri chment analysis.The main biological pathways include HIF-1 signaling pathway a nd endocrine resistance pathway,etc.Finally,the results of molecular docking show that there is a strong affinity between the key active components and the t arge.CONCLUSION The results showed that Bupleurum-Scutellaria might play the role of“Treating different diseases together”by regulating i nflammatory reaction and improving gastrointestinal function.
