基于数据挖掘从“方证病位”探讨血府逐瘀汤

摘　　要：目的通过数据挖掘和生物信息学技术探讨王清任“血府”的科学内涵,并深入探索血府逐瘀汤的临床应用规律,有利于扩大血府逐瘀汤的临床用药范围。方法通过中国国家知识基础设施(www.cnki.net,CNKI)、万方数据(www.wanfangdata.com.cn,Wanfang Data)、维普网(www.cqvip.com,VIP)和中国生物医学文献服务系统(www.sinomed.ac.c,CBMdisc)对含血府逐瘀汤进行检索,应用TCMISS(V2.5)、Excel行频数分析和聚类分析。运用网络药理学技术,于HERB数据库下载血府逐瘀汤11味药物的作用靶点。利用R软件的clusterProfiler包进行疾病本体(DO)富集分析。结果共筛选包含血府逐瘀汤医案107个,涉及辅助中药183种,使用频率较高的中药依次为黄芪、白芍、丹参、郁金。药性多为平、温、微寒;药味偏苦、辛、甘;归经多归肝、心、脾经。利用TCMISS(V2.5)软件的“新方分析”功能得到3个核心方剂。通过疾病富集分析得出集中于心血管、呼吸、泌尿等系统疾病,符合血府的部位位于胸中的推测。结论血府逐瘀汤疏肝结行气滞而不伤阴,主治胸中血瘀诸证,是活血祛瘀和调理气血的常用方剂,体现了中医“异病同治”的指导思想。OBJECTIVE To explore the scientific connotation of“Xuefu”in Wang Qingren through data mining and bioinformatics technology,and to deeply e xplore the clinical application law of XueFu ZhuYu Decoction,which is conducive to expanding the clinical application scope of XueFu ZhuYu Decoction.METHODS The XueFu ZhuYu Decoction was searched through the National K nowledge Infrastructure of China(www.cnki.net,CNKI),Wanfang Data(www.wanf angdata.com.cn,Wanfang Data),Weipu.com(www.cqvip.com,VIP)and China Bio medical Literature Service System(www.sinomed.ac.cn,CBMdisc),and the frequ ency analysis and cluster analysis were performed by using TCMISS(V2.5)and Exc el.Using network pharmacology technology,the target of 11 drugs of XueFu ZhuYu Decoction was downloaded from HERB database.The clusterProfiler package of R s oftware was used for disease(DO)enrichment analysis.RESULTS A total of 107 medical records containing XueFu ZhuYu Decoction were screened,i nvolving 183 kinds of auxiliary Chinese medicines,and the most frequently used Chinese medicines were Astragalus membranaceus,Paeonia lactiflora,Salvia milti orrhiza and Radix Curcumae.Most of the drugs are flat,warm and slightly cold;The medicine tastes bitter,pungent and sweet;Meridian tropism mostly belongs t o liver,heart and spleen meridians.Three core prescriptionswere obtained by us ing the“new prescription analysis”function of TCMISS(V2.5)software.Throug h the disease enrichment analysis,it is concluded that the diseases concentrate d in cardiovascular,respiratory and urinary systems are consistent with the spe culation that the blood house is located in the chest.CONCLUSION XueFu ZhuYu Decoction is a common prescription for promoting blood circulati on,removing blood stasis and regulating qi and blood,which embodies the guidin g ideology of“treating different diseases at the same time”in traditional Ch inese medicine.
