摘 要:【目的】筛选蓝莓花粉败育相关基因,为其克隆、蓝莓花粉败育研究和育种提供参考依据。【方法】用Illumina NovaSeq 6000对来源于相同母本且花粉活力有显著差异的‘灿烂(BT)’‘杰兔(PM)’和‘粉蓝(PB)’3个兔眼蓝莓品种花粉的转录组进行测序,基于差异表达基因的基因本体数据库(GO)、Pathway显著性富集(KEGG)和蛋白家族数据库(Pfam)分析,筛选参与蓝莓花粉败育的相关基因。【结果】‘杰兔’花粉活力显著低于‘灿烂’和‘粉蓝’,‘灿烂’vs‘杰兔’与‘粉蓝’vs‘杰兔’2个对比组合分别筛选到2717和3208个差异表达基因。‘灿烂’vs‘杰兔’对比组合的2717个差异表达基因在GO、KEGG和Pfam数据库中分别已注释1124、1020和1791个。‘粉蓝’vs‘杰兔’对比组合的3208个差异表达基因在GO、KEGG和Pfam数据库中分别已注释1350、1144和2197个。对差异表达基因的GO和Pfam注释结果进一步筛选,获得与花粉败育相关的差异表达基因27个,其中,22个上调表达,5个下调表达。【结论】经GO和Pfam注释,花粉败育相关差异表达基因功能主要集中在花粉发育、花穗发育、孢粉素生物合成过程、花粉外壁形成、花药壁绒毡层细胞分化、细胞分裂素的正向调节、有丝分裂纺锤体组织调节和油菜素内酯介导信号途径等通路,可作为深入探索蓝莓花粉败育机制及筛选蓝莓花粉败育关键基因的参考依据。[Objective]The genes related to pollen abortion of blueberry were screened to provide reference for its cloning and related research and breeding.[Method]Illumina NovaSeq 6000 was used to sequence the transcriptomes of pollen from three rabbiteyed blueberry cultivars,‘Brightwell(BT)',‘Powderblue(PB)'and‘Premier(PM)',which were derived from the same female parent and had significant differences in pollen viability.The genes related to pollen abortion of blueberry were screened based on the Gene Ontology(GO),Kyoto Encyclopedia of Genes and Genomes(KEGG)and Protein family(Pfam).[Result]The pollen viability of‘PM'was significantly lower than that of‘BT'and‘PB',and 2717 and 3208 differentially expressed genes were detected in the comparison combinations of‘BT'vs‘PM'and‘PB'and‘PM',respectively.There were 1124,1020 and 1791 differentially expressed genes in the GO,KEGG and Pfam databases of the 2717 differentially expressed genes in the comparison combination of‘BT'vs‘PM',while the 3208 differentially expressed genes of‘PB'vs‘PM'were annotated 1350,1144 and 2197 differentially expressed genes in GO,KEGG and Pfam databases,respectively.Further screening of GO and Pfam annotation results of differentially expressed genes revealed that 27 differentially expressed genes were related to pollen abortion,of which 22 genes were up-expressed and 5 genes were down-expressed.[Conclusion]The annotation results of GO and Pfam differentially expressed genes related to pollen abortion show that pollen development,inflorescence development,sporopollenin biosynthetic process,formation of pollen exine,cell differentiation of anther wall tapetum,positive regulation of cytokinesis,regulation of mitotic spindle organization,brassinosteroid mediated signal process and other pathways are closely related to pollen abortion in blueberry.It can be used as a reference for further exploring the mechanism of pollen abortion and mining the key genes of pollen abortion in blueberry.
