摘  要:为探究小麦COL基因在生长发育过程中的潜在功能,以中国春为研究对象,采用生物信息学方法在小麦全基因组水平鉴定TaCOL基因并进行系统分析,并利用qRT-PCR检测其在不同组织及不同胁迫条件下的表达。结果显示,在小麦全基因组水平上一共鉴定到48个COL基因,在进化关系上被分为Ⅰ、Ⅱ、Ⅲ三组;亚细胞定位预测表明,所有TaCOL蛋白均定位在细胞核中。染色体定位发现,它们不均等地分布在小麦的16条染色体上,其中有4个TaCOL基因组成了4个片段复制基因对。基因结构和保守基序分析发现,相同亚组的TaCOL在基因结构和保守基序组成上类似,其中Motif 1和Motif 2分别是构成B-box和CCT两个保守结构域的主要基序。此外,在TaCOL基因的启动子区分布着许多光调节、激素响应、生长发育等多种类型顺式作用元件。qRT-PCR分析发现,所有的TaCOL基因在小麦的不同组织中均有表达,且大部分基因在叶片中的表达水平较高;多数TaCOL基因响应非生物胁迫和激素的诱导,不同光周期处理也能显著影响TaCOL基因的表达水平。本研究为进一步研究TaCOL基因在小麦的生长发育和作物遗传改良方面奠定了基础。To explore the potential function of COL genes in the growth and development of wheat,bioinformatics methods were used to systematically analyze and identify TaCOL genes at the whole-genome level of wheat with“Chinese spring”as material.The expression patterns of these genes under different tissue types,stress conditions,and light and dark treatments were detected by qRT-PCR.The results showed that a total of 48 COL genes were identified in the whole-genome level of wheat,which were divided into three groups ofⅠ,Ⅱ,andⅢin evolutionary relationships.Subcellular localization prediction indicated that all TaCOL proteins are located in the nucleus.Chromosome localization analysis showed that they are unevenly distributed on 16 chromosomes of wheat,and 4 TaCOL genes formed 4 fragment pairs of replication genes.Gene structure and conserved motif analysis found that the TaCOL genes in the same subgroup had similar gene structures and conserved motif compositions,and Motif 1 and Motif 2 were the main motifs that constitute the B-box and CCT conserved domains,respectively.Additionally,many cis-acting elements of five types,including light regulation,hormone response,growth and development,stress response,and other types,were found in the promoter regions of TaCOL genes.Expression pattern analysis showed that all TaCOL genes were expressed in different tissues of wheat,and most genes had higher expression levels in leaves.Most TaCOL genes responded to abiotic stress and hormone induction,and long day or short day treatment significantly affected the expression levels of TaCOL genes.This study lays a foundation for further research on the function of TaCOL genes in wheat growth and development and crop genetic improvement.
