小麦金属硫蛋白全基因组鉴定及镉胁迫下的表达分析

摘  要:为探究小麦金属硫蛋白(metallothionein,MT)基因的潜在功能,以小麦品种中国春、M1019和西农20为材料,采用生物信息学方法,在小麦全基因组水平系统分析和鉴定了TaMT基因,并利用转录组数据分析和qRT-PCR技术检测在镉胁迫条件下,小麦不同组织和不同品种中的表达。结果显示,在小麦全基因组水平上共鉴定到21个TaMT基因,他们在进化关系上被分为Ⅰ、Ⅱ、Ⅲ三组。TaMT蛋白可定位在细胞不同位置,暗示这些基因可能在不同部位发挥功能。经染色体定位分析,这些基因不均等地分布在小麦的8条染色体上,其中4个TaMT基因形成了4个串联复制基因对。此外,相同亚组的TaMT在基因结构和保守基序组成上表现出相似性,其中motif 3构成了α结构域,而motif 1和motif 4组成了β结构域。在TaMT基因的启动子区分布着多种类型的顺式作用元件,包括植物发育、激素响应、胁迫响应、光调节和其他类型。表达分析表明,TaMT基因在小麦不同组织中均能检测到,同时镉胁迫能够诱导TaMT基因在不同品种和小麦组织中发生差异表达。To investigate the potential functions of metallothionein(MT)genes in wheat,this study employed bioinformatics methods to systematically analyze and identify TaMT genes at the whole-genome level.Transcriptome data and qRT-PCR were utilized to detect their expression patterns in different tissues and wheat varieties under cadmium stress.The results showed that a total of 21 TaMT genes were identified across the wheat genome,and divided into three subgroups(Ⅰ,Ⅱ,Ⅲ)based on their evolutionary relationships.Subcellular localization prediction indicated that TaMT proteins are located in different sections of the cell,suggesting that they may function in different compartments.Chromosomal localization analysis revealed that they are unevenly distributed on the eight chromosomes of wheat,with four TaMT genes forming four tandem duplicated gene pairs.Gene structure and conserved motif analysis showed that TaMTs within the same subgroup exhibited similarity in gene structure and conserved motif composition,with motif 3 constituting theα-domain and motif 1 and motif 4 composing theβ-domain.Additionally,various types of cis-acting elements were identified in the promoter regions of TaMT genes,including those related to plant development,hormone response,stress response,light regulation,and other types.Expression pattern analysis demonstrated that TaMT genes were detectable in different tissues of wheat,and cadmium stress induced differential expression of TaMT genes in different wheat varieties and tissues.
