摘  要:目的:优化异体垂体移植术诱导小鼠子宫腺肌病模型,为相关研究提供稳定、便捷的动物模型。方法:在前人工作基础上,将供体垂体混合匀浆并采用胰岛素注射器定量注射入雌鼠子宫腔,分别在造模16周(w)和24 w后处死小鼠,测量子宫质量及系数,HE染色检测子宫组织病理,ELISA法测定血清泌乳素含量。结果:优化的造模方法能够降低供体垂体个体差异和操作误差,减少受体子宫腔损伤。造模16 w和24 w后,优化的造模方法能够成功诱导小鼠典型的子宫腺肌病病理现。结论:本研究建立的造模方法,可为子宫腺肌病发病机制和药物开发研究提供更为稳定、便捷的动物模型。Objective:To optimize the mouse model of adenomyosis induced by allogeneic pituitary transplantation,and provide a stable and convenient animal model for related research.Methods:Based on previous work,this study mixed the homogenate of the donor pituitary and injected quantitatively into the uterine cavity of female mouse by insulin syringe.The mice were sacrificed at 16 weeks and 24 weeks after modeling.The uterine mass and coefficient was measured.The uterine tissue histopathology was detected by HE staining,and the serum prolactin content was determined by ELISA method.Results:The optimized modeling method could reduce the individual difference of the donor pituitary,minimize operation error and reduce the injury of the recipient uterine cavity.The typical pathological characteristics of adenomyosis were successfully reproduced at 16weeks and 24 weeks after modeling.Conclusion:The modeling method established in this study can provide a more stable and convenient animal model for the pathogenesis and drug development of adenomyosis.
