摘  要:多环芳烃(PAHs)污染物的可降解性能较低,在自然环境长期蓄积,造成环境污染及生物毒害作用。苍白杆菌(Ochrobactrum sp.) FP1是从石油污染土壤分离的高效石油降解菌。采用涂布平板计数、高效液相色谱(HPLC)等方法研究了该菌在含菲基础培养基中的生长特性及最适生长条件,探究了葡萄糖、甘油、邻苯二酚、水杨酸和鼠李糖脂等不同外加基质存在下菌体的生长特性及菲降解能力。结果表明,苍白杆菌FP1能在p H5~8和20~40℃的条件下,在菲含量为50 mg·L^(-1)的基础培养基中正常生长,其最适生长pH 7、最适温度为30℃。外加基质葡萄糖、甘油及鼠李糖脂对菌株生长有良好促进作用,甘油的促进作用最明显,而水杨酸和邻苯二酚对菌株的生长有抑制作用。HPLC分析结果显示,外加基质甘油、葡萄糖、鼠李糖脂处理组的降解率分别较空白组提升了21.58%、15.38%、6.76%,而水杨酸和邻苯二酚处理组的降解率分别下降了46.27%和70.31%,细菌对菲降解率的变化与其生长能力呈正相关,高浓度葡萄糖对菌株代谢菲有抑制作用,苍白杆菌FP1对菲的降级酶系为诱导酶。本研究结果可为细菌共代谢多环芳烃提供理论参考。Polycyclic aromatic hydrocarbons(PAHs)have low degradability and accumulate in natural environment for a long time,causing environmental pollution and biological toxicity.Ochrobactrum sp.FP1 is an efficient petroleum degrading bacterium isolated from oil contaminated soil.The growth characteristics and optimum growth conditions of Ochrobactrum sp.FP1 were studied by coated plate counting and High Performance Liquid Chromatography(HPLC).The growth characteristics and phenanthrene degradation ability of bacteria in the presence of different external substrates such as glucose,glycerol,catechol,salicylic acid and rhamnoolipid were studied.The results showed that Ochrobactrum sp.FP1 could grow normally at pH 5~8 and 20~40℃ with phenanthrene content of 50·mg L^(-1),and the optimum growth temperature was 30℃ and pH 7.The addition of substrates glucose,glycerol and rhamnose-lipids had a good promotion effect on the growth of the strain,and glycerol was the most obvious promotion effect,while salicylic acid and catechol had an inhibitory effect on the growth of the strain.The results of HPLC analysis showed that compared with the control group,the degradation rates of the additional substrates glycerol,glucose and rhamnose-grease treatment groups were increased by 21.58%,15.38%and 6.76%,respectively,while the degradation rates of salicylic acid and catechol treatment groups were decreased by 46.27%and 70.31%,respectively.The change of bacterial degradation rate of phenanthocyanin was positively correlated with its growth ability.High glucose concentrations inhibited the metabolism of phenanthrene by the strain,and the degradation enzymes of phenanthrene in Ochrobactrum sp.FP1 were inducible enzymes.The results of this study can provide theoretical reference for bacterial co-metabolism of polycyclic aromatic hydrocarbons.
