摘  要:黄瓜是全世界栽培最广、生产和消费最多的蔬菜作物之一,具有较高的经济价值。为了加强黄瓜种质资源的保护与利用,本试验利用259份黄瓜材料的重测序数据进行SNP鉴定,根据位点缺失率、杂合度以及多态性信息含量等指标对SNP集合进行筛选,最终获得了能够高效鉴定黄瓜种质的150个核心SNP标记。该套核心SNP位点组合的PIC和MAF平均值分别为0.374、0.479。通过聚类分析可知,259份黄瓜种质可以被划分为东亚型和欧亚型两大类群。基于以上核心SNP标记为每一份黄瓜种质构建了DNA指纹图谱,能够有效区分所有供试材料。本研究为黄瓜遗传多样性分析、种质资源鉴定以及新品种培育提供了重要的理论依据。Cucumber is one of the most widely cultivated,produced,and consumed vegetable crops in the world,with high economic value.In order to strengthen the protection and utilization of cucumber germplasm resources,this experiment used resequencing data from 259 cucumber materials for SNP identification.The SNP set was screened based on indicators such as site deletion rate,heterozygosity,and polymorphism information content.Finally,150core SNP markers that can efficiently identify cucumber germplasm were obtained.The average PIC and MAF values of the core SNP site combination in this set are 0.374 and 0.479,respectively.Through cluster analysis,it can be concluded that 259 cucumber germplasms can be divided into two major groups:East Asian type and Eurasian type.A DNA fingerprint map was constructed for each cucumber germplasm based on the core SNP markers mentioned above,which can effectively distinguish all the tested materials.This study provides important theoretical basis for genetic diversity analysis,germplasm resource identification,and new variety cultivation of cucumber.
