摘  要:探究无糖培养条件下,不同因素对黑金刚莲雾扦插的影响,并筛选出最优的无糖扦插组合,为莲雾的扦插育苗提供理论基础。利用无糖组织培养设备,以黑金刚莲雾枝条为插穗,设置营养液种类(A)、枝条类型(B)、生根剂ABT溶液浓度(C)及浸泡时间(D)四因素三水平的正交试验,并在此基础上进行优化试验。结果表明:正交试验中对莲雾生根效果的影响大小为A>D>C>B,最优组合为A_(1)B_(3)C_(1)D_(2);对生根率的影响大小为B>A>D>C,最优组合为A_(1)B_(2)C_(1)D_(2);对腐烂率的影响大小为C>A>D>B,最优组合为A_(3)B_(1)C_(1)D_(2)。优化试验以MS(无糖)营养液+蛭石为扦插基质,插穗在浓度为150或250 mg/L的ABT溶液中浸泡2 min时,扦插效果指数分别为2.90、2.83,此处理有利于黑金刚莲雾的枝条生根,扦插效果较好。To investigate the effects of different factors on the cutting of the‘Black Diamond’Syzygium samarangense Merr.et Perry.under sugar-free culture conditions,and select the optimal sugar-free cutting combination,providing a theoretical basis for the cutting and seedling cultivation of lotus mist.This study utilized sugar-free tissue culture equipment,using'Black Diamond'S.samarangense Merr.et Perry.branches as cuttings and conducted orthogonal experiments at four factors and three levels:nutrient solution type(A),branch type(B),rooting agent ABT solution concentration(C),and soaking time(D).Fur-ther optimization experiments were conducted on this basis.The results revealed that the factors influencing the rooting effect of lotus mist in the orthogonal experiments were A>D>C>B,and the optimal combination was A_(1)B_(3)C_(1)D_(2).The factors affecting the rooting rate were B>A>D>C,and the optimal combination was A_(1)B_(2)C_(1)D_(2).The influencing factors of the decay rate are C>A>D>B,and the optimal combination is A_(3)B_(1)C_(1)D_(2).The optimization experiment used MS(sugar-free)nutrient solution and vermiculite as the cutting substrate.When soaked in ABT solution at concentrations of 150 or 250 mg×L-1 for 2 minutes,the cutting effect indices were 2.90 and 2.83,respectively.This treatment is conducive to rooting the branches of‘Black Dia-mond’Syzygium samarangense Merr.et Perry.and the cutting effect is good.
