摘  要:本研究采用两步乳化法制备葡聚糖-铁蛋白-白藜芦醇纳米粒子(dextran-rHuHF-resveratrol,Dex-rHuHFRes)稳定的W1/O/W2多重乳液,并对多重乳液进行表征,分析其稳定性及对生物活性物质的递送作用。结果表明,在聚甘油蓖麻醇酸酯添加量为7%(以油相体积计)、W1与O体积比为3∶7、W1/O初乳与外水相W2体积比为4∶6的条件下,乳液外观均一、液滴分布均匀、粒径最小(472 nm),表观黏度和储能模量(G’)最高。在pH 7、NaCl浓度100 mmol/L、低离心力条件下乳液稳定性较好。在不利环境中,与游离的绿原酸、虾青素及白藜芦醇相比,多重乳液所负载的绿原酸、虾青素及白藜芦醇保留率显著提升(P<0.05)。W1/O/W2多重乳液负载的绿原酸、虾青素及白藜芦醇的生物可及性显著升高(P<0.05),乳液的抗氧化活性明显提升(P<0.05)。本研究结果可为Dex-rHuHF-Res纳米颗粒稳定的W1/O/W2多重乳液应用于递送活性物质提供理论参考。In this study,a water-in-oil-in-water(W1/O/W2)multiple emulsion stabilized by dextran-ferritin-resveratrol(Dex-rHuHF-Res)nanoparticles was prepared by a two-step emulsification method.Its stability and potential for the delivery of bioactive substances were analyzed.The results showed that when the concentration of polyglycerol ricinoleate(PGPR)was 7%(V/V),the ratio of aqueous phase(W1)to oil phase(O)was 3:7,and the ratio of W1/O to outer aqueous phase(W2)was 4:6,the emulsion had homogeneous appearance,uniform droplet size distribution,the smallest particle size(472 nm),and the highest apparent viscosity and storage modulus(G’).At pH 7,100 mmol/L NaCl concentration and low centrifugal force,the emulsion maintained good stability.In unfavorable environment,compared with free chlorogenic acid,astaxanthin and resveratrol,the retention rates of chlorogenic acid,astaxanthin and resveratrol loaded in the multiple emulsion were significantly improved(P<0.05).The bioaccessibilities of chlorogenic acid,astaxanthin,and resveratrol loaded in the W1/O/W2 emulsion were significantly increased(P<0.05),and the antioxidant property of the emulsion was obviously enhanced(P<0.05).This study provides a theoretical basis for the application of the W1/O/W2 emulsion stabilized by Dex-rHuHF-Res nanoparticles in the delivery of active substances.
