摘  要:为明确呈现褪绿和花叶症状的南京萝卜的具体病原,应用反转录PCR法对采集的萝卜病样进行了14种病毒的检测及序列分析.结果表明:南京萝卜病样感染了黄瓜花叶病毒(cucumber mosaic virus,CMV)和芜菁花叶病毒(turnip mosaic virus,TuMV),进一步对该萝卜样品中扩增到的CMV外壳蛋白基因(coat protein,CP)序列分析发现,CMV CP基因序列与已公开的苦瓜、烟草和茄子等分离物的CP序列高度同源,序列相似性高达98%,系统进化树表明CMV CP序列属于ⅠB亚组;该萝卜病样的TuMV CP序列与来自韩国、朝鲜及日本等国家的TuMV分离物CP序列同源性最高,相似性达99.65%;系统进化树表明,TuMV CP序列属于TuMV的Basal-BR亚组.研究结果鉴定了南京地区萝卜病样感染的病毒种类,通过CP基因序列分析明确了感染南京萝卜的CMV与TuMV与其他分离物之间的亲缘关系,为制定相应的防控措施提供了理论基础.In order to clarify the specific pathogens of diseased Nanjing radishes with chlorosis and mosaic symptoms,14 viruses were detected by RT-PCR.The amplified results showed that diseased Nanjing radish plants were infected with cucumber mosaic virus(CMV)and turnip mosaic virus(TuMV).In addition,the coat protein(CP)nucleic acid sequences of CMV and TuMV were isolated and sequenced.Sequence alignment analysis showed that the CMV CP sequences isolated from Nanjing radish were highly homologous with those of bitter gourd,tobacco and eggplant,and the identities of nucleic acid sequences were higher than 98%.Phylogenetic analysis using CMV CP sequence showed that the CMV CP sequence isolated from Nanjing radish belonged to the IB subgroup.The TuMV CP sequence of Nanjing radish had the high homology with those of radish isolated from South Korea,the Democratic People′s Republic of Korea,Japan and other regions of China,and the highest similarity was 99.65%.Phylogenetic analysis showed that the TuMV CP sequence i-solated from Nanjing radish belonged to the Basal-BR subgroup.The results identified the viruses infected the radish plants Nanjing,and clarified the genetic relationship between the identified viruses and other isolates through sequence analysis of CP gene.The analysis findings could provid a theoretical basis for prevention and control measures for radish virus diseases.
