摘  要:研究旨在建立高效液相色谱法(HPLC)同时测定公英青蓝合剂中黄芩苷和绿原酸含量的方法。公英青蓝合剂经20%乙醇提取后,以C18色谱柱(4.6 mm×150 mm,3.5μm)为分离柱,柱温为35℃,检测波长为324 nm,二极管阵列检测器,外标法定量。结果表明:黄芩苷在20.91~209.12 mg/L和绿原酸在5.12~51.20 mg/L范围内线性关系良好(R2>0.9990);黄芩苷平均加样回收率为101.09%,变异系数为1.15%;绿原酸平均加样回收率为99.59%,变异系数为1.46%;系统适应性试验良好,色谱柱耐受性良好,完全满足检测需求。研究表明,试验所用方法可操作性强,稳定性、重复性、准确性良好,适用于公英青蓝合剂中黄芩苷和绿原酸含量的同时检测。The aim of this study was to establish a HPLC method for the simultaneous determination of baicalin and chlorogenic acid in Gongyingqinglan Heji.After extraction with 20% methanol,Gongyingqinglan Heji was separated by C18 chromatographic column(4.6 mm×150 mm,3.5μm).The column temperature was set at 35℃,the detection wavelength was 324 nm,the photo-diode array detector was used for analysis,and external standard method was used for quantification.The results showed that the linear relationship between baicalin and chlorogenic acid was good within the ranges of 20.91~209.12 mg/L and 5.12~51.20 mg/L,respectively(R2>0.9990).The average recovery rate of baicalin was 101.09%,and the coefficient of variation was 1.15%.The average recovery rate of chlorogenic acid was 99.59%,and the coefficient of variation was 1.46%.The system adaptability test was good,and the chromatographic column had good tolerance,fully meeting the detection requirements.The study indicates that this method has strong operability,with good stability,repeatability and accuracy,and is suitable for the simultaneous determination of baicalin and chlorogenic acid content in Gongyingqinglan Heji.
