摘 要:目的:观察辛麻颗粒对慢性哮喘小鼠气道上皮屏障功能的影响。方法:将40只BALB/C雌性小鼠按随机数字表法分成对照组、哮喘组、辛麻颗粒组(辛麻组)、地塞米松组(地米组),每组10只。采用卵清白蛋白(OVA)致敏和激发建立慢性哮喘小鼠模型。苏木素-伊红(HE)染色观察肺组织病理学改变;酶联免疫吸附法(ELISA)测定支气管灌洗液白细胞介素-25(IL-25)、白细胞介素-33(IL-33)的水平;免疫组化及免疫荧光法检测肺组织E钙黏蛋白(E-cadherin);RT-PCR测定RORαmRNA的表达。结果:HE染色可见哮喘组小鼠气道炎症明显。哮喘组小鼠支气管灌洗液IL-25、IL-33水平高于对照组(P<0.05),辛麻组、地米组IL-25、IL-33水平低于哮喘组(P<0.05)。免疫组化法和免疫荧光法检查结果显示哮喘组小鼠肺组织E-cadherin蛋白相对表达量低于对照组(P<0.05),辛麻组和地米组肺组织E-cadherin蛋白表达高于哮喘组(P<0.05)。与对照组比较,哮喘组小鼠肺组织RORαmRNA的表达增强(P<0.05);与哮喘组比较,辛麻组、地米组小鼠肺组织RORαmRNA的表达下降(P<0.05)。结论:辛麻颗粒能够减轻慢性哮喘小鼠气道炎症及改善气道上皮屏障功能,为其临床应用治疗哮喘提供实验依据。Objective:To observe the effect of Xinma Granules on the airway epithelial barrier function in mice with chronic asthma.Methods:A total of 40 female BALB/C mice were randomly divided into the control group,the asthma group,the Xinma Granules group(Xinma group),and the dexamethasone group(DXM group),with 10 mice in each group.The chronic asthma mice models were established by sensitization and stimulation with ovalbumin(OVA).Pathological changes in lung tissue were observed using hematoxylin eosin(HE)staining method;enzyme linked immunosorbent assay(ELISA)was used to measure the levels of interleukin-25(IL-25)and interleukin-33(IL-33)in bronchoalveolar lavage fluid;immunohistochemical and immunofluorescence methods were used to detect E-cadherin in lung tissue;RT-PCR was used to measure the expression of RORαmRNA.Results:HE staining showed significant airway inflammation in the asthma group.The levels of IL-25 and IL-33 in bronchial lavage fluid in the mice of the asthma group were higher than those in the control group(P<0.05),and the levels of IL-25 and IL-33 in the Xinma group and DXM group were lower than those in the asthma group(P<0.05).Immunohistochemical and immunofluorescence methods found that the relative expression of E-cadherin protein in lung tissue in the asthma group was lower than that in the control group(P<0.05).The expression of E-cadherin protein in lung tissue of the Xinma group and DXM group was higher than that in the asthma group(P<0.05).Compared with the control group,the expression of RORαmRNA in lung tissue of the asthma group was enhanced(P<0.05);compared with the asthma group,the expression of RORαmRNA in lung tissue of mice in the Xinma group and DXM group was decreased(P<0.05).Conclusion:Xinma Granules can alleviate airway inflammation and improve airway epithelial barrier function in mice with chronic asthma,providing experimental evidence for their clinical application in the treatment of asthma.
