摘  要:【目的】应用rbcL叶绿体序列解析金线兰及其近缘种亲缘关系。【方法】以血叶兰属及开唇兰属rbcL叶绿体序列设计引物,扩增福建主栽金线兰及其近缘种的DNA序列,并进行系统进化树及序列分析。【结果】从7种金线兰及其近缘种中获得1221 bp rbcL序列,编码406个氨基酸。对来自19种23条金线兰及近缘种rbcL序列的进化树及序列分析结果表明,金线兰与峨眉金线兰、长裂片金线兰及浙江开唇兰有较近的遗传距离,其次为台湾银线兰,血叶兰与开唇兰属存在较大的遗传距离。23条序列共存在24个多态性位点,及109个变异位点,定义14个单倍型;其中,金线兰与峨眉金线兰、长裂片金线兰及浙江开唇兰共享单倍型,而丽蕾金线兰、滇南开唇兰、滇越金线兰、白线金线兰、钟山金线兰共享单倍型。【结论】rbcL序列可用于金线兰与近缘种的鉴别。【Objective】Based on rbcL chloroplast sequence analysis to determine the phylogenetic relationships between Anoectochilus roxborghil and its related species.【Method】Primers were designed based on the rbcL chloroplast sequences of Ludisia and Anoectochilus genera.Sequences were amplified from the DNA of the main cultivated A.roxborghil and its related species in Fujian,followed by phylogeneticand sequence analysis.【Result】rbcL sequences with 1221 bp and coding 406 amino acids were amplified from 7 A.roxborghil and its related species.Total 19 species and 23 rbcL sequences were using for phylogenetic analysis.Results revealed that A.roxborghil was closely related to A.emeiensis,A.longilobus,and A.zhejiangensis,followed by A.formosanus.And there is a significant genetic distance between Ludisia and Anoectochilus genera.Those 23 sequences generated a total of 24 polymorphic sites and 109 variant sites,defined 14 haplotypes.Among them,A.roxborghil shared same haplotype with A.emeiensis,A.longilobus,and A.zhejiangensis,while A.lylei,A.burmannicus,A.chapaensis,A.albolineatus,A.zhongshanensis shared same haplotype.【Conclusion】rbcL sequences could be used to distinguish between A.roxborghil and its related species.
