摘  要:目的:探究海藻糖敷料对激光损伤兔动物模型皮肤的修复作用,并探索其体外促细胞生长、迁移和抗炎作用。方法:试验兔脊柱两边各脱毛4个区域,随机分为正常组(N)、模型组(M)、对照组(P)和实验组(T)。除正常组外,其余各组使用激光照射造模;对照组及实验组分别使用透明质酸敷料及海藻糖敷料贴敷。于第0天、第3天、第7天、第14天和第21天对创面进行红斑和焦痂、水肿程度评分,以评价海藻糖敷料对皮肤修复效果。以L929细胞及HaCaT细胞为研究对象探索海藻糖敷料体外促细胞生长、迁移和抗炎作用。将细胞分为空白组(培养基)、正常组(细胞液+培养基)、对照组(细胞液+培养基+透明质酸)及实验组(细胞液+培养基+海藻糖)。使用MTT比色法检测各组L929细胞24 h、48 h及72 h增殖率,划痕试验检测各组L929细胞6 h、12 h和24 h迁移力;使用TNF-α及IFN-γ诱导HaCaT细胞建立炎症反应模型后,24 h后检测各组IL-6、IL-8和MDC表达水平。结果:创面给予敷料后7 d,实验组水肿评分[(1.1±0.3)分]较模型组[(1.2±0.4)分]显著降低(P<0.05);第14天,相较于模型组,实验组及对照组水肿、红斑焦痂评分有下降趋势但差异无统计学意义(P>0.05)。病理结果显示,对照组、实验组较模型组第14天炎细胞浸润微有减少。细胞实验结果显示,与正常组相比,对照组和实验组4 h、48 h及72 h的细胞增殖率均有明显增加;与正常组相比,对照组及实验组6 h、12 h和24 h的细胞迁移率均有明显增加(P<0.05);与细胞炎症模型组相比,实验组细胞上清液中IL-6、IL-8和MDC的表达水平明显降低(P<0.01)。结论:海藻糖敷料对激光所致兔皮肤屏障受损创面的组织炎症、愈合有一定修复作用;其机制可能与其能促进细胞增殖、细胞迁移及抑制炎症的功能相关。Objective To evaluate the effects of trehalose dressing on skin repair after laser surgery in rabbits,and to explore its effects on cell proliferation,migration and anti-inflammatory in vitro.Methods Four areas of hair were removed on each side of the spine in rabbits,which were randomly divided into normal group(N),model group(M),control group(P)and experimental group(T).Except for the normal group,rabbits in the rest of the groups were irradiated by laser.The control group and the experimental group was treated by hyaluronic acid dressing and trehalose dressing respectively.To evaluate the effect of trehalose dressings on skin repair,erythema,eschar and edema was scored on day 0,3,7,14,21.L929 cells and HaCaT cells were used to explore effects of trehalose on cell proliferation,migration and anti-inflammatory in vitro.Cells were divided into blank group(cell culture medium,CCM),normal group(cell+CCM),control group(cell+CCM+hyaluronic acid)and experimental group(cell+CCM+trehalose).The growth of L929 cells was assayed by MTT method at 24h,48h and 72 h.The migration of L929 cells was assyed by cell scratch experiment at 6 h,12 h and 24 h.Level of IL-6,IL-8 and MDC was tested after TNF-αand IFN-γinduced inflammation in HaCaT cells after 24h.Results On the 7th day,the edema score(1.1±0.3)in the experimental group was significantly lower than that in the model group(1.2±0.4)(P<0.05).On the 14th day,the edema,erythematous and eschar score of the experimental group and the control group had a downward trend compared with the model group,but there were no significant difference(P>0.05).The pathological results showed that,on the 14th day,the control group and the experimental group were slightly reduced in inflammatory cell infiltration compared with the model group.Results of cell experiment showed that compared with the normal group,proliferation rate of L929 cells was significantly increased in control group and experimental group at 24 h,48 h and 72 h(P<0.05).Compared with the normal group,migration rate of L92
