摘  要:探讨海南省红带锥蝽中感染锥虫的分子流行特征,为锥虫病的防治提供依据。2020—2021年在海南省采集红带锥蝽,设计属通用引物,采用PCR扩增锥虫18S小亚基核糖体RNA基因(18S rDNA),电泳阳性结果测序,阳性率的比较采用卡方检验。对阳性样本进行18S rDNA和热休克蛋白70基因(HSP70)序列比对和遗传变异分析。2020—2021年在海南昌江黎族自治县和五指山市共采集锥蝽143只,经实验室鉴定均为红带锥蝽。锥虫18S rDNA扩增,共获得51份锥虫阳性样本,锥虫阳性率为35.66%,其中五指山和昌江地区红带锥蝽锥虫阳性率分别为52.00%和26.88%。对51份18S rDNA测序分析,结果表明:海南省红带锥蝽感染锥虫分为两类,一类与Trypanosoma conorhini 18S rDNA(XR_003828665.1)序列一致性高达100.00%;另一类与T.conorhini(XR_003828665.1)序列一致性为95.79%左右,为T.conorhini地理株。对两类阳性样品进行HSP70扩增克隆测序获得9条阳性样品HSP70的核酸序列。构建系统发育树,结果表明:T.conorhini分为两群,一群与T.conorhini(XR_003828665.1,AJ012411.1)亲缘关系更近,同为一个分支;另一群单独聚为一支。本次对海南省昌江、五指山两地红带锥蝽的调查显示:T.conorhini野生型阳性率为6.29%,T.conorhini地理株的阳性率为29.37%。本次调查的海南地区红带锥蝽感染的锥虫均为T.conorhini,并出现了T.conorhini地理株,且其为优势虫株。To investigate the molecular epidemiological characteristics of Trypanosoma infection in Triatoma rubrofasciata in Hainan Province,providing a basis for the prevention and control of trypanosomiasis.T.rubrofasciata were collected in Hainan Province from 2020 to 2021.General primers were designed,and PCR was used to amplify the Trypanosoma 18S small subunit ribosomal RNA gene(18S rDNA).Positive products were sequenced.Chi-square test was used to compare the positive rates.Genetic variation and sequence comparison of the 18S rDNA and the heat shock protein 70 gene(HSP70)were conducted for positive samples.Confi rmed by laboratory identifi cation,a total of 143 T.rubrofasciata were collected in Changjiang Li Autonomous County and Wuzhishan City,Hainan Province,from 2020 to 2021.The 18S rDNA gene of trypanosomes was amplifi ed,resulting in 51 trypanosome-positive samples,with a positivity rate of 35.66%.The positive rates of Trypanosoma infection in T.rubrofasciata were 52.00%in Wuzhishan and 26.88%in Changjiang.Sequence analysis of the 18S rDNA gene from 51 positive samples revealed that Trypanosoma infections in T.rubrofasciata in Hainan Province are divided into two groups:one group shows 100.00%sequence identity with T.conorhini 18S rDNA(XR_003828665.1),while the other shows a sequence identity of about 95.79%with T.conorhini(XR_003828665.1),indicating a geographic strain of T.conorhini.Cloning and sequencing of the HSP70 from positive samples identifi ed 9 nucleotide sequences.Phylogenetic analysis showed that T.conorhini can be divided into two groups:one closely related to T.conorhini(XR_003828665.1,AJ012411.1)and another consisting of geographic strains of T.conorhini.The investigation of T.rubrofasciata in Changjiang and Wuzhishan revealed a wild-type T.conorhini positivity rate of 6.29%and a geographic strain positivity rate of 29.37%respectively.Trypanosoma infection in T.rubrofasciata in Hainan Province are predominantly caused by T.conorhini,including a geographic strain that appears to be the dominant s
