摘 要:以分离株PEDV/Belgorod/dom/2008的全基因组序列为研究对象,利用MUSCLE软件对GenBank数据库中的18个全基因组PEDV序列、3个传染性胃肠炎病毒(transmissible gastroenteritis virus,TGEV)序列和1个猪肠道冠状病毒序列进行多重比对,通过MEGA 6.0中的极大似然法,基于PEDV M和S基因序列构建系统发育树。结果检测到PEDV/Belgorod/dom/2008的S基因重组事件,全基因组序列两两同一性分析发现,PEDV/Belgorod/dom/2008是PEDV与传染性胃肠炎病毒毒株之间的中间体。表明PEDV/Belgorod/dom/2008是一种新的重组毒株,PEDV/Belgorod/dom/2008与已知的PEDV毒株关系较远。PEDV/Belgorod/dom/2008分离株不属于美国或中国毒株形成的任何类群,并与意大利分离的SeCoV-ITA09重组毒株形成单独的集群。此项研究可用于进一步分析猪流行性腹泻病毒的进化变异性、流行率和流行病学。The complete genome sequences of the PEDV/Belgorod/dom/2008 strain were used to perform multiple alignment of 18 genome-wide PEDV sequences,3 Transmissible gastroenteritis virus(TGEV)sequences and one Porcine intestinal coronavirus sequence in the GenBank database using MUSCLE software.Phylogenetic trees were constructed based on PEDV M and S gene sequences by maximum likelihood method in MEGA 6.0.The results showed that the recombination event was detected in S gene of PEDV/Belgorod/dom/2008 and PEDV/Belgorod/dom/2008 was an intermediate between PEDV and transmissible gastroenteritis virus(TGEV)strains based on pairwise identity analysis of the complete genome sequence.These results indicated that PEDV/Belgorod/dom/2008 was a novel recombinant strain and distantly related to known PEDV strains.The PEDV/Belgorod/dom/2008 strain did not belong to any group consisting of the US or Chinese strains and formed a separate cluster with the SeCoV-ITA09 recombinant strain isolated in Italy.The information from this study could be used to further analyze the evolutionary variability,prevalence and epidemiology of PEDV.
