摘 要:细胞周期调控与细胞增殖和分化、细胞凋亡和癌变密切相关。这种调节涉及多种信号传导途径,它们的正常进行对维持细胞周期的稳定性非常重要。然而,信号通路中蛋白质的水平和/或活性会受到多种影响,导致细胞周期失调,最终可能导致癌症的发生。因此,阐明导致细胞周期失调的特定信号通路对癌症治疗非常重要。蛋白质泛素化在调控包括细胞周期在内的各种生理过程中发挥着重要作用,通过调控关键蛋白质的表达或活性来影响细胞周期。USP家族的一个重要成员USP22被证明在多种人类癌症中上调,并与Cyclin D1、Cyclin B1、BMI-1、MYC、p53及其他细胞周期调控的重要因子相互作用,导致细胞周期进展的改变。本文以USP22相关的细胞周期调控因子为重点,进一步阐述了USP22是如何调控细胞周期并导致肿瘤发生的,并总结了USP22在癌细胞周期中发挥调控作用的途径,这将为后续肿瘤治疗药物的开发提供思路。Cell cycle regulation is closely related to cell proliferation and differentiation,apoptosis and carcinogenesis.This regulation involves multiple signaling pathways,the proper conduct of which is important for maintaining cell cycle stability.However,the levels and/or activities of proteins in the signaling pathways can be affected in a variety of ways,resulting in cell cycle dysregulation,which may ultimately lead to cancer development.Therefore,elucidating the specific signaling pathways that cause cell cycle dysregulation is important for cancer treatment.Protein ubiquitination plays an important role in the regulation of various physiological processes,including cell cycle,which is affected by ubiquitination through the regulation of the expression or activity of key proteins.Recently,USP22,an important member of the USP family,has been shown to be up-regulated in a variety of human cancers and interacts with Cyclin D1,Cyclin B1,BMI-1,MYC,p53 and other important factors in cell cycle regulation,leading to alterations in cell cycle progression.In this paper,focusing on USP22-related cell cycle regulators,we further explain how USP22 regulates the cell cycle and causes tumorigenesis,and summarize the pathways through which USP22 plays a regulatory role in the cancer cell cycle,which will provide ideas for the subsequent development of oncology therapeutic drugs.
