摘  要:以柠檬酸铁和聚乙烯亚胺(polyethyleneimine,PEI)为初始原料,采用微波水热法,快速合成了具有良好光致发光性能的铁掺杂碳点(Fe doped carbon dots,Fe-CDs).研究表明:在较宽的pH范围(pH=3~9)、较高的离子强度和较长的储存时间下,Fe-CDs表现出优异的稳定性,易穿透细胞膜,并在细胞内聚集,可作为肺癌A549细胞生物成像的有效探针;Fe-CDs具有优异的类过氧化物酶(peroxidase-like,POD)活性,能够高效催化过氧化氢(H_(2)O_(2))对3,3′,5,5′-四甲基联苯胺(TMB)的氧化,并生成蓝色氧化产物,基于此建立了H_(2)O_(2)比色测定方法;在最佳反应条件下,H_(2)O_(2)浓度为0~100.00μmol/L时,H_(2)O_(2)与反应体系的吸光度呈现良好的线性关系,H_(2)O_(2)检出限低至0.13μmol/L,说明该比色测定方法对H_(2)O_(2)的检测表现出较高的灵敏度和选择性;将该比色测定方法用于A549细胞内H_(2)O_(2)的测定,加样回收率高达98.4%~102.8%,验证了该方法的可靠性.The novel Fe doped carbon dots(Fe-CDs) with good photoluminescent properties were rapidly synthesized by microwave-assisted method with ferric citrate and polyethyleneimine as raw materials.The results show that the Fe-CDs have good stability under the conditions of broad pH range from 3 to 9,high ion strengths and long storage time.The prepared Fe-CDs can easily penetrate and accumulate into the cell,which can be used as effective probe for biological imaging of A549 cells.The Fe-CDs display distinct peroxidase-like(POD) activities,which is evidenced by the catalyzed oxidation of 3,3′,5,5′-tetramethylbenzidine(TMB) into blue oxidized products in the presence of H_(2)O_(2).A colorimetric method for the determination of H_(2)O_(2) is established based on the results.Under optimal reaction conditions,the colorimetric method for H_(2)O_(2) detection is constructed with good linear relationship from 0 to100.00 μmol/L and detection limit down to 0.13 μmol/L.The highly selective and ultrasensitive method is successfully applied to detect H_(2)O_(2) inside A549 tumor cells with good recoveries of 98.4%-102.8%,which can verify the proposed method.
