香薷水提液对急性热应激小鼠血常规、血清生化和抗氧化功能的影响

摘  要:试验旨在探讨香薷水提液对热应激小鼠血常规、血清生化及抗氧化功能的影响,将30只ICR小鼠随机分为空白组、模型组、阳性对照组及香薷水提液低、中、高剂量组。空白组和模型组灌胃生理盐水,阳性对照组给予太极藿香正气口服液,香薷水提液低、中、高剂量组分别灌胃0.5、1.0、2.0 mL/kg的香薷水提液,均按照2 mL/kg体重灌胃,每日1次,连续7 d。最后一次给药结束后进行热应激试验,置于40℃恒温箱中热应激2 h后检测小鼠血常规指标、血清生化指标、抗氧化相关指标及热休克蛋白70(HSP70)指标。结果表明:与模型组相比,香薷水提液能显著降低急性热应激小鼠白细胞、中性粒细胞数量(P<0.05),显著降低血清天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、HSP70及肝脏丙二醛(MDA)水平(P<0.05),显著提高肝脏总超氧化物歧化酶(SOD)、谷胱苷肽过化物酶(GSH-Px)活性(P<0.05),以高剂量组效果最好。综上,香薷水提液可缓解急性热应激所致小鼠血液炎性细胞升高,改善热应激小鼠肝、肾损伤及抗氧化功能,降低血清HSP70的含量,提高机体的抗热应激能力,以高剂量2.0 mL/kg效果最佳。The experiment aimed to explore the effects of Elsholtzia ciliata aqueous extract on the blood tests,serum biochemistry and antioxidant function of heat-stressed mice.Thirty ICR mice were randomly divided into blank group,model group,positive control group,and different dose groups(low,medium and high)of Elsholtzia ciliata aqueous extract.The blank group and model group were administered physiological saline by gavage,the positive control group was given Huoxiangzhengqi oral liquid,and the low,medium,and high dose groups of Elsholtzia ciliata aqueous extract were respectively administered 0.5,1.0,and 2.0 mL/kg of the extract by gavage,all at a dose of 2 mL/kg BW,once daily for 7 days.Af⁃ter the final administration,heat stress test was conducted by placing the mice in a 40℃constant tem⁃perature box for 2 hours to detect the blood routine parameters,serum biochemical parameters,antioxidant-related parameters,and heat shock protein 70(HSP70)levels.The results showed that com⁃pared with the model group,the Elsholtzia ciliata aqueous extract significantly reduced the white blood cell and neutrophil counts of mice under acute heat stress(P<0.05),significantly decreased the levels of serum aspartate aminotransferase(AST),alanine aminotransferase(ALT),HSP70,and liver malondialdehyde(MDA)(P<0.05),increased the levels of liver total superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)(P<0.05),with the highest efficacy observed in the high dose group.In conclusion,the water extract of Elsholtzia ciliata aqueous extract can alleviate the increase of in⁃flammatory cells in the blood of mice induced by acute heat stress,improve the liver,kidney,and antioxi⁃dant function of heat-stressed mice,reduce the content of serum HSP70,enhance the body′s anti-heat stress ability,with the best effect observed in the high dose group of 2.0 mL/kg.
