摘 要:目的:探讨猫爪草总皂苷(TSRT)基于Sema4D/PlxnB1/c-Met通路对人非小细胞肺癌A549裸鼠移植瘤增殖的抑制作用及机制。方法:成功构建A549裸鼠移植瘤模型后,随机分为空白组、阴性组、过表达组和药物组(1.73g/kg TSRT),记录成瘤情况并绘制肿瘤生长曲线,实验结束后剥离移植瘤并称重;RT-PCR和Western blot分别检测各组Sema4D、PlxnB1、c-Met mRNA和蛋白表达差异;免疫组化检测Sema4D阳性表达率。结果:与空白组相比,过表达组接种20d后的肿瘤体积显著增加,肿瘤重量明显增加,Sema4D、PlxnB1、c-Met mRNA和蛋白表达水平显著升高,Sema4D阳性表达率显著提高(P<0.01);与过表达组相比,药物组接种20d后肿瘤体积明显降低,瘤重显著降低,Sema4D、PlxnB1、c-Met mRNA和蛋白表达水平显著下降,Sema4D阳性表达率明显降低(P<0.05或P<0.01)。结论:TSRT能显著抑制人肺癌裸鼠移植瘤的生长,其作用机制可能与抑制Sema4D/PlxnB1/c-Met信号通路有关。Objective:To explore the inhibitory effect and mechanism of total saponins of Ranunculus ternatus(TSRT)on the proliferation of A549 transplanted tumor in nude mice based on Sema4D/PlxnB1/c-Met path⁃way.Methods:After model of A549 transplanted tumor in nude mice were successfully constructed,they were randomLy divided into blank group,negative group,overexpression group and drug group(gavaged with 1.73g.kg-1 TSRT).Recorded the tumor formation and drawn the tumor growth curve,after the experiment,the transplanted tumor was peeled off and the tumor weight was weighed.RT-PCR and Western blot were used to detect differences in Sema4D,PlxnB1,and c-Met mRNA and protein expression in each group.The posi⁃tive expression rate of Sema4D was detected by immunohistochemistry.Results:Compared with the blank group,the tumor volume of 20 days after inoculation in the over-expression group were significantly higher,and the tumor weight increased significantly,the expression levels of Sema4D,PlxnB1,c-Met mRNA and pro⁃tein increased significantly,and the positive expression rate of Sema4D increased significantly(P<0.01).Com⁃pared with the over-expression group,the tumor volume of 20 days after inoculation in the drug group were significantly lower,and the tumor weight decreased significantly,the expression levels of Sema4D,PlxnB1,cMet mRNA and protein decreased significantly,and the positive expression rate of Sema4D decreased signifi⁃cantly(P<0.05 or P<0.01).Conclusion:TSRT can significantly inhibit the growth of human lung cancer transplanted tumor in nude mice,and its mechanism may be related to the inhibition of Sema4D/PlxnB1/cMet signaling pathway.
