摘 要:为阐明鸭防御素在禽流感病毒(Avian influenza virus,AIV)引起的免疫反应中的作用,研究首先分析转录组数据,选定研究对象后构建过表达载体,并将过表达载体转染至DEF或DF-1细胞,随后用H5N1亚型AIV感染细胞,通过测定病毒滴度进行研究。结果显示:鸭防御素3(Avian beta defensins 3,AvBD3)在鸭感染两株H5N1亚型AIV后表达量均上升,其中AvBD3C上调最为显著;在AvBD3C及其突变体mAvBD3C过表达的细胞中,H5N1亚型AIV滴度显著降低,表明鸭AvBD3C及其突变体mAvBD3C均可以抑制H5N1亚型AIV的复制。研究表明,鸭AvBD3C具有抗H5N1亚型AIV活性。In order to explore the role of duck defensins in the immune response induced by avian influenza virus(AIV),the study firstly analyzed the transcriptome data and selected the object after which overexpression vectors were constructed.The vectors were transfected into DEF or DF-1 cells,and then the cells were infected with H5N1 subtype AIV.The results showed that the expression of duck avian beta defensins 3(AvBD3)was up-regulated in ducks infected with two strains of H5N1 subtypes AIV,and the up-regulation of AvBD3C was the most significant.In the cells overexpressed AvBD3C and mAvBD3C,the titers of H5N1 subtype AIV were significantly decreased,indicating that both duck AvBD3C and its mutant mAvBD3C could inhibit the proliferation of H5N1 subtype AIV.The results indicated that duck AvBD3C had anti-H5N1 AIV activity.
