摘 要:试验旨在调查H9N2亚型禽流感病毒(Avian influenza virus,AIV)在江苏省常州市的流行情况。2023年3—12月,在常州市所有辖区的交易平台、散养户、活禽市场、屠宰场等7种场所类型的147个采样场点开展H9N2亚型AIV流行病学调查,试验采集鸡、鸭、鹅、鸽等咽拭子、组织样品以及环境拭子共计1970份,进行AIV分离及其流行病学特征分析,并对分离的H9N2亚型AIV进行遗传演化、关键氨基酸与糖基化位点和抗原性分析。结果显示:共鉴定出73株H9N2亚型AIV,平均分离率为3.70%;4月和5月分离率较高,分别为7.07%和8.00%;分离地点中活禽市场和交易平台的分离率最高;H9N2亚型AIV在鸡、鸭、鸽、野鸟粪便和环境中均有分离,在环境和鸡中的分离率最高,分别为6.90%和4.15%;分离的H9N2亚型AIV属于欧亚谱系9.1分支中Group2群,与2021—2023年多个地区人感染H9N2亚型AIV亲缘关系较近;与参考株序列相比,H9N2亚型AIV分离株HA蛋白的130-loop、150-loop及190-helix存在明显氨基酸多态性,但没有发现新的糖基化位点;H9N2亚型AIV分离株均属于当前优势流行的抗原群,尚未发生明显抗原漂移。综上所述,江苏常州地区H9N2亚型AIV流行的热点场所是活禽市场和交易平台,提示环境消毒是控制病毒传播和污染的关键手段;H9N2亚型AIV可能在野生禽类中流行,须减少家禽与野禽接触;并且H9N2亚型AIV分离株HA蛋白受体结构域变异可能对人间感染造成持续威胁,值得关注。To investigate the prevalence of H9N2 subtype avian influenza virus(AIV)in Changzhou,Jiangsu province,an epidemiological survey was conducted from March to December 2023.A total of 1970 samples including throat swabs from chickens,ducks,geese and pigeons,tissue samples and environmental swabs were collected across 147 sampling sites encompassing 7 administrative areas of Changzhou at trading platforms,free-range households,live poultry markets and slaughterhouses.AIV was isolated from collected samples and epidemiological characteristics was analyzed;The isolated H9N2 subtype AIVs were further analyzed for genetic evolution,key amino acid variations and antigenicity.The results showed that there were 73 strains of H9N2 subtype AIVs with an average isolation rate of 3.70%in this study.The highest isolation rates were recorded in April and May,at 7.07%and 8.00%,respectively.The highest isolation rates were found in live poultry markets and trading platforms.H9N2 subtype AIVs were isolated from chickens,ducks,pigeons,wild bird feces and the environment,with the highest rates in environmental samples and chickens,at 6.90%and 4.15%,respectively.The H9N2 subtype AIVs belonged to the Eurasian lineage 9.1,Group 2 cluster,and were closely related to the strains infecting humans in various regions from 2021 to 2023.There were significant polymorphisms in the HA protein of H9N2 subtype AIV isolates at the 130-loop,150-loop and 190-helix,and no new glycosylation site was found compared to the reference strains.The H9N2 subtype AIV isolates belonged to the currently dominant antigenic group with no significant antigenic drift observed.In conclusion,live poultry markets and trading platforms were the hotspot venue types for the prevalence of H9N2 subtype AIV in Changzhou,which emphasized that environmental disinfection was crucial for controlling virus transmission and contamination.The prevalence of H9N2 subtype AIV in wild birds suggested a need to reduce contact between domestic and wild birds.Variations in the receptor-bin
