摘 要:猪繁殖与呼吸综合征(Porcine reproductive and respiratory syndrome,PRRS)是一种严重威胁猪群健康的病毒性疾病。为进一步了解猪繁殖与呼吸综合征病毒(Porcine reproductive and respiratory syndrome virus,PRRSV)HLJ-80株GP5蛋白的结构和功能,对该病毒株的GP 5基因进行了生物信息学和序列分析。结果显示,HLJ-80株GP5蛋白编码201个氨基酸,基因全长为603 bp,该蛋白分子质量为22.2 kDa,等电点(pI)为8.85,不稳定系数为39.17。该病毒株GP5蛋白属于疏水性蛋白,含有信号肽,其剪切位点位于第31~32位氨基酸之间。亚细胞定位预测显示GP5蛋白定位于内质网中。根据跨膜螺旋结构的预测,GP5蛋白存在3个跨膜螺旋结构。此外,该蛋白存在38个磷酸化位点和29个O-糖基化位点。抗原性结果显示,GP5蛋白的柔韧性较差,表面可及性区域较少。氨基酸序列同源性结果表明,HLJ-80株与NADC30株的GP5蛋白同源性最高,与GM2株的GP5蛋白同源性最低,且HLJ-80株GP5蛋白存在9个氨基酸突变位点。基因系统结果显示,HLJ-80株与NADC30株GP 5基因具有最近的遗传距离,与QYYZ株GP 5基因具有最远的遗传距离,本研究为监测PRRSV的流行特点提供新的数据。Porcine reproductive and respiratory syndrome is viral disease that threatens the health of swime.In order to further understand the structure and function of GP5 protein of porcine reproductive and respiratory syndrome virus(PRRSV)HLJ-80 strain,the GP5 gene of this strain was analyzed by bioinformaties and sequence analysis.The results showed that the GP5 protein of HLJ-80 strain encoded 201 amino acids,the total length of the gene was 603 bp,the molecular weight of the protein was 22.2kDa,the isoelectric point(pI)was 8.85,and the instability coefficient was 39.17.The GP5 protein of this strain is a hydrophobic protein,containing a signal peptide,and its shear site is between the 31 and 32 amino acids.Subcellular localization prediction showing GP5 protein results was partially displayed in the endoplasmic reticulum.According to the prediction of transmembrane helical structure,GP5 protein has three transmembrane helical structures.In addition,the protein has 38 phosphorylation sites and 29 O-glycosylation sites.The antigenicity results showed that GP5 protein had poor flexibility and fewer surface accessible areas.Amino acid sequence homology showed that the GP5 protein homology of HLJ-80 and NADC30 was the highest,and the GP5 protein homology of GM2 was the lowest,and there were 9 amino acid mutation sites in GP5 protein of HLJ-80.The results of gene system showed that HLJ-80 had the closest genetic distance from NADC30 and the farthest genetic distance from QYYZ.This study provided new data for monitoring the epidemic characteristics of PRRSV.
