摘 要:基于胶原中苯丙氨酸与酪氨酸产生可定量内源性荧光的原理,利用同步荧光光谱和发射荧光光谱研究了牦牛皮胶原质量浓度对其自组装行为的影响及牦牛皮胶原组装过程中的荧光共振能量转移现象,并通过计算确定0.5 mol/L乙酸中牦牛皮胶原的临界组装质量浓度为0.77 mg/mL。建立了牦牛皮胶原荧光强度与0.10~3.00 mg/mL质量浓度的对应工作曲线,并由此对胶原通过0.22μm过滤膜的效率进行了定量分析,证明通过率与质量浓度的自然对数呈线性关系。牦牛皮胶原在酸性无盐条件下形成复杂的自组装结构,而中性盐的加入改变了组装状态和进程,在磷酸缓冲液-氯化钠的存在下,自组装效果最佳。随着组装程度增加,胶原逐渐形成有序的空间结构,吸光度、荧光强度、模量和复合黏度上升,损耗系数降低。Based on the principle that phenylalanine and tyrosine in collagen produce quantitative endogenous fluorescence,the effect of yak hide collagen mass concentration on its self-assembly behavior and the phenomenon of fluorescence resonance energy transfer during the assembly process were studied by synchronous fluorescence spectrum and emission fluorescence spectrum,and the critical assembly mass concentration of yak hide collagen in 0.5 mol/L acetic acid was determined to be 0.77 mg/mL by calculation.The corresponding working curve between fluorescence intensity of yak hide collagen and its mass concentration of 0.10-3.00 mg/mL was established,and the efficiency of collagen passing through O.22μm filter membrane was quantitatively analyzed,which proved that the passing rate was linear with the natural logarithm of mass concentration.Yak hide collagen formed the complex self-assembly structure under acidic saltfree conditions,and the addition of neutral salt changed the assembly state and process.In the presence of phosphate buffer-sodium chloride,the self-assembly effect is optimum.With the increase of assembly degree,collagen gradually formed the orderly spatial structure,and the absorbance,fluorescence intensity,modulus and complex viscosity increased,while the loss coefficient decreased.
