摘  要:本研究以苹果砧木平邑甜茶(Malus hupehensis var.pingyiensis)cDNA为模板,克隆出bHLH家族成员MhbHLH122;该基因编码区全长1314 bp,编码蛋白具有bHLH保守结构域,与桃PpbHLH122转录因子亲缘关系最近。将MhbHLH122转入‘王林’苹果愈伤组织,获得了MhbHLH122过表达的愈伤株系;在含200 mmol·L^(–1)NaCl的培养基上,过表达MhbHLH122的愈伤组织比野生型愈伤组织生长状态更好,其丙二醛(MDA)含量、细胞相对死亡率、过氧化氢(H_(2)O_(2))含量、超氧阴离子(O_(2)^(¯·))产生速率均低于野生型,超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性均高于野生型,显示过表达MhbHLH122能增强苹果愈伤组织耐盐性。This paper used the cDNA of apple rootstock Malus hupehensis var.pingyiensis as a template,cloned MhbHLH122,a member of bHLH family which has a total length of 1314 bp gene coding region,its encoded protein contains a bHLH conserved domain and shares the highest similarity with the peach PpbHLH122 transcription factor.Transferring MhbHLH122 into calli of‘Orin’apple resulted in the overexpression of MhbHLH122 in calli.On a culture medium containing 200 mmol·L^(–1)NaCl,the growth status of calli overexpressing MhbHLH122 was better than that of the wild-type,the malondialdehyde(MDA)content,relative cell death rate,hydrogen peroxide(H_(2)O_(2))content,and superoxide anion(O_(2)^(¯·))production rate were all lower than in the wild-type,the activities of superoxide dismutase(SOD),peroxidase(POD),and catalase(CAT)were all higher than the wild-type,in the overexpressed MhbHLH122 compared to the wildtype,suggested that the overexpression of MhbHLH122 can improve salt tolerance in apple calli.
