摘 要:为了解新疆地区某猪场猪嗜血支原体的感染情况,试验采集新疆地区某猪场保育仔猪的抗凝血200份,采用血液压片、血涂片瑞氏染色镜检和普通PCR方法检测猪嗜血支原体,并利用猪嗜血支原体16SrRNA基因引物,以血液DNA为模板,扩增、克隆、鉴定后送测序,进行同源性比对与遗传进化分析。结果显示,经血液压片、血液涂片及PCR检测共检出58份猪嗜血支原体阳性样本;以猪嗜血支原体16SrRNA基因引物、血液DNA为模板成功克隆猪嗜血支原体基因,命名为XJP1(已被Genbank收录,登录号:PP526316)。同源性比对与遗传进化分析结果显示,XJP1与猪嗜血支原体(登录号:KC907396)、猪嗜血支原体(登录号:HQ259257)为同一分支,亲缘关系较近,同源性为96.9%~99.6%。研究表明,该猪场保育猪感染了猪嗜血支原体,试验结果将为该猪场防控猪嗜血支原体感染以及该病的深入研究提供基础数据。In order to understand the infection situation of Mycoplasma suis in a pig farm in Xinjiang region,200 samples of anticoagulant blood of piglets in a pig farm in Xinjiang region were collected.Blood pressure tablet,blood smear staining microscopy and common PCR method were used to detect Mycoplasma suis.16S rRNA gene primer of Mycoplasma suis was used,and blood DNA was used as template.After amplification,cloning,identification and sequencing,homology comparison and genetic evolution analysis were performed.The results showed that 58 positive samples of Mycoplasma suis were detected by blood pressure smear,blood smear and PCR.Using the 16S rRNA gene primer of Mycoplasma suis and blood DNA as the template,the gene of Mycoplasma suis was successfully cloned and named XJP1(has been included by Genbank,entry number:PP526316).Homology comparison and genetic evolution analysis showed that XJP1 was the same cladus with Mycoplasma suis(entry number:KC907396)and Mycoplasma suis (entry number:HQ259257),and the homology was 96.9%~99.6%.The study showed that the piglets in the farm were infected with Mycoplasma suis,and the test results will provide basic data for the prevention and control of Mycoplasma suis infection in the farm and the further study of the disease.
