摘 要:文中选用冷水鱼皮明胶(CFG)为基材,通过定量修饰饱和脂肪链癸醛(C10)及儿茶酚基团3,4-二羟基苯甲醛(CAT)制备了3种改性明胶,再将改性明胶与四臂聚乙二醇琥珀酰亚胺戊二酸酯(4arm-PEG-NHS)混合交联,构建了具有高黏弹性的组织密封胶。通过伯胺基团的测定和紫外光谱表征,确定了C10及CAT改性基团的取代度。双重改性明胶(CAT-C10-CFG)与4arm-PEG-NHS交联成水凝胶时间短,可在5 s内迅速成胶,具有良好的组织黏附性和伸缩性能。在体外模型评价中发现,CAT-C10-CFG密封胶的耐压爆破强度可达87.1 mmHg,远超肺部所承受的局部生理压力,表现出优异的肺组织密封性能。细胞毒性评价中,L-929细胞的存活率达到95%以上,表明密封胶材料具有良好的生物相容性,在胸外科领域具有良好的应用前景,为术后肺漏气的堵漏治疗提供了新思路。Cold-water fish skin gelatin(CFG)was selected as substrate.Three modified gelatins were prepared by quantitatively modifying saturated fat chains capraldehyde(C10)and catechol groups 3,4-dihydroxybenzalde-hyde(CAT).Then,the modified gelatin was mixed and cross-linked with a four-arm polyethylene glycol succinimi-dyl glutarate(4arm-PEG-NHS)to construct a tissue sealant with high viscoelasticity.The degree of substitution of C10 and CAT modified groups was determined by the determination of primary amine groups and ultraviolet(UV)spectroscopy.The double-modified gelatin(CAT-C10-CFG)and 4arm-PEG-NHS were cross-linked into hydrogels quickly,which could be gelatinized within 5 s,and the hydrogels have good tissue adhesion and elastic properties.In the evaluation of in vitro model,it is found that the pressure burst strength of CAT-C10-CFG sealant could reach 87.1 mmHg,which far exceeds the local physiological pressure of the lungs and shows excellent lung tissue seal-ing performance.In the cytotoxicity evaluation,the survival rate of L-929 cells reaches more than 95%,indicating that the sealant material has a good biocompatibility.Given this,CAT-C10-CFG tissue sealant has a broad applica-tion prospect in thoracic surgery,which provides a new idea for treating postoperative pneumothorax leakage.
