摘 要:目的探讨紫檀芪对人舌鳞癌CAL-27细胞生物学行为的影响。方法将CAL-27细胞分为对照组和实验组,实验组用紫檀芪10、20、40μmol/L进行处理。CCK-8法、克隆形成实验和EdU检测不同浓度紫檀芪对CAL-27细胞增殖的影响;流式细胞术检测CAL-27细胞凋亡情况;划痕实验和穿孔实验实验观察CAL-27细胞的迁移和侵袭能力;蛋白质印迹检测CAL-27细胞内上皮间质转化(epithelial-mesenchymal transition,EMT)相关蛋白的表达。异种移植瘤验证紫檀芪对体内肿瘤的作用。结果与对照组比较,紫檀芪20和40μmol/L处理24 h后CAL-27细胞增殖、迁移和侵袭能力减弱,细胞凋亡率增加,细胞内Bax、E-cadherin和Claudin1蛋白表达量增加,Bcl-2、N-cadherin、波形蛋白和Snail蛋白的表达量减少(P<0.05)。紫檀芪30 mg/kg治疗后裸鼠体内移植肿瘤重量和体积明显减小,Ki67和波形蛋白阳性细胞数明显减少,TUNEL阳性细胞数明显增加(P<0.05)。结论紫檀芪能抑制CAL-27细胞增殖、迁移、侵袭和EMT,同时诱导其凋亡。Objective Explore the effect of Pterostilbene on the biological behavior of human tongue squamous cell carcinoma CAL-27 cells.Methods CAL-27 cells were divided into 2 groups:control group and experimental group.Cells in the experimental group were treated with Pterostilbene(10,20,40μmol/L).CCK-8 assay,colony formation assay and EdU staining were used to measure the proliferation of CAL-27 cells.Flow cytometry was used to determine the apoptosis of CAL-27 cells.Wound-healing and Transwell assay were used to detect the migration and invasion ability of CAL-27 cells.Western blotting was used to detect the expression levels of epithelial-mesenchymal transition(EMT)-related proteins in CAL-27 cells.Xenograft model was used to verify the effect of pterostilbene on tumor in vivo.Results The proliferation,migration and invasion abilities of CAL-27 cells were weakened,the apoptosis rate was increased,the expression levels of Bax,E-cadherin and Claudin1 proteins in the cells were increased,and the expression levels of Bcl-2,N-cadherin,Vimentin and Snail proteins were decreased after 24 h treatment with 20 and 40μmol/L pterostilbene when compared with those in the control group(P<0.05).After treatment with 30 mg/kg pterostilbene,the weight and volume of transplanted tumors in nude mice decreased significantly,the number of Ki67 and Vimentin positive cells decreased significantly,and the number of TUNEL positive cells increased significantly(P<0.05).Conclusion Pterostilbene can inhibit CAL-27 cells proliferation,migration,invasion and EMT and induce its apoptosis.
