重齿毛当归(武陵山独活)提取物的抗氧化、舒缓、修护功效研究

摘　　要：重齿毛当归(武陵山独活)根可入药,功效与作用主要是祛风湿、解表、止痛。本论文采用乙醇/水加热萃取的工艺提取了叶、花原料,并研究多种提取物在抗氧化、舒缓和修护方面的性能。当提取物浓度为2.5 mg/mL时,“花-35乙醇”和“叶-35乙醇”样品的羟基自由基清除率达到89.45%和74.93%;当提取物浓度为6.25 mg/mL时,“叶-100水”和“花-100水”样品的透明质酸酶抑制率为74.94%和61.53%;当提取物浓度为2 mg/mL时,“花-85乙醇”、“叶-100水”、“叶-85乙醇”和“花-100水”样品的人永生化表皮细胞增值率分别为121.61%、122.58%、118.98%和119.41%;此外,提取物样品对小鼠胚胎成纤维细胞的细胞毒性极小或者无。研究表明重齿毛当归(武陵山独活)的叶和花提取物具有显著的抗氧化活性,能够有效的清除羟基自由基,抑制透明质酸酶的活性,增加皮肤的保湿能力,并减轻皮肤炎症反应,还能促进人永生化表皮细胞的增殖,以达到修护皮肤和延缓衰老的功能并且没有毒性或毒性极小。为重齿毛当归(武陵山独活)的综合利用提供借鉴。The root of ANGELICA PUBESCENS(Wuling Mountain Angelica) has been used in traditional Chinese medicine for dispelling rheumatism,relief epilepsy and alleviating pain.The extracts from the leaves and flowers of ANGELICA PUBESCENS(Wuling Mountain Angelica) were evaluate in the antioxidant,soothing,and repairing.The hydroxyl radical scavenging rates of the “Flower-35 ethanol” and “Leaf-35 ethanol” samples were 89.45% and 74.93%,respectively,at a concentration of 2.5 mg/mL.When the concentration of the extract was 6.25 mg/mL,the hyaluronidase inhibition rates of the “Leaf-100 water” and “Flower-100 water” samples were 74.94% and 61.53%,respectively.At a concentration of 2 mg/mL,the cell proliferation rates of the “Flower-85 ethanol”,“Leaf-100 water”,“Leaf-85 ethanol” and “Flower-100 water” samples in immortalized human epidermal cells were 121.61%,122.58%,118.98% and 119.41%,respectively.Furthermore,the extract samples exhibited minimal or no cytotoxicity towards mouse embryonic fibroblast cells.The extracts of ANGELICA PUBESCENS(Wuling Mountain Angelica) leaves and flowers showed significant antioxidant activity,effectively scavenge hydroxyl radicals,inhibiting hyaluronidase activity.They could also enhance the skin moisturization,reduce skin inflammation,and promote the proliferation of immortalized human epidermal cells.Moreover,these extracts exhibited skin repairing and anti-aging properties with minimal or negligible toxicity.This research provided valuable insights for the comprehensive utilization of ANGELICA PUBESCENS(Wuling Mountain Angelica).
