摘 要:目的观察去氢骆驼蓬碱(HAM)是否对多器官功能障碍综合征(MODS)大鼠肺组织炎症损伤具有保护作用。方法构建MODS动物模型,大鼠随机分为对照组、MODS组和HAM组。检测肺湿干重比值、肺系数,HE染色观察肺组织形态,采用ELISA试剂盒检测血清白细胞介素-6(IL-6)、IL-10和肿瘤坏死因子-α(TNF-α)的水平,免疫组织化学染色检测肺组织S100钙结合蛋白A8/A9(S100 A8/A9)的定位表达,Western blotting法检测肺组织S100 A8/A9、M1型巨噬细胞标志物诱导型一氧化氮合酶(iNOS)和M2型巨噬细胞标志物精氨酸酶1(Arg-1)的蛋白表达。结果与MODS组相比,HAM组肺湿干重比值、肺系数减小,肺泡腔炎性渗出减少,肺间质充血水肿减轻,血清IL-6、TNF-α水平降低,IL-10水平升高,肺组织S100 A8/A9、iNOS表达下降,Arg-1表达升高。结论HAM可通过抑制炎症反应降低MODS大鼠肺组织损伤。Objective To observe the protective effect of harmine(HAM)on lung tissue inflammation injury in rats with multiple organ dysfunction syndrome(MODS).Methods MODS animal model was established and rats were randomly divided into control group,MODS group and HAM group.The lung wet-dry weight ratio and lung coefficient were calculated respectively.Lung tissue morphology was observed by HE staining.The levels of IL-6,IL-10 and TNF-αin serum were detected by ELISA kit.The localized expression of S100 calcium-binding protein A8/A9(S100 A8/A9)was detected by immunohistochemical staining.Western blotting was used to detect the expression of S100 A8/A9,M1 macrophage marker inducible equivalent of nitric oxide synthase(iNOS),and M2 macrophage marker arginase-1(Arg-1)in lung tissue.Results Compared with MODS group,the ratio of lung wet and dry weight and lung coefficient in HAM group were decreased,inflammatory exudation of alveolar cavity was reduced,pulmonary interstitial hyperemia and edema were alleviated,serum IL-6 and TNF-αlevels were decreased,IL-10 levels were increased,S100 A8/A9 and iNOS expression in lung tissue were decreased,and Arg-1 expression was increased.Conclusion HAM can improve lung tissue injury in MODS rats by inhibiting inflammatory response.
