摘 要:为探究瓠瓜(Lagenaria siceraria)CAT基因家族的特性及在白粉病菌侵染后的作用,基于瓠瓜基因组数据和转录组数据,对LsCATs家族成员进行了鉴定与分析,研究其表达模式。结果表明:在瓠瓜基因组中共鉴定到2个CAT家族同源基因,其编码蛋白氨基酸数分别为489、1019个,且均为亲水性蛋白;进化分析表明,瓠瓜CAT基因家族被分成2个亚组,推测在相同亚组的瓠瓜与黄瓜CAT转录因子可能具有相似的功能;利用MEME软件预测得到15个motif元件,不同CAT基因家族的motif数量及分布存在一定差异;顺式作用元件分析表明,LsCATs的启动子包含ABA、乙烯、MeJA、干旱、低温等多种与逆境胁迫有关的响应元件;瓠瓜接种白粉病菌后,LsCAT1表达量呈现出先升高后降低的趋势,LsCAT2则表现为先下降后升高的趋势,表明LsCAT1、LsCAT2可能与瓠瓜的白粉病抗性相关。本研究可为CAT基因在瓠瓜抗病机制的研究提供理论依据。To investigate the characteristics of CAT family in Lagenaria siceraria and its roles after powdery mildew infection,this study identified and analyzed LsCATs members and its expression patterns using bioinformatics software based on the genomic data and transcriptome data of Lagenaria.The results showed that two CAT family genes were identified in bottle gourd genome,with an amino acid number of 489 and 1019,respectively,all of which were hydrophilic proteins;Evolutionary tree analysis showed that LsCAT was divided into two subgroups,indicating that bottle gourd and cucumber CAT transcription factors in the same subgroup might have similar functions;15 motif elements were predicted using MEME software.Differences were existed in motif numbers and locations among different CATs.The analysis of cis acting elements indicated that the promoter of LsCATs contained ABA,ethylene,and MeJA,drought,and other stess-related elelments.Analysis of the expression levels of LsCATs after inoculation with powdery mildew pathogen in bottle gourd revealed that LsCAT1 showed a trend of first increasing and then decreasing,while LsCAT2 showed a trend of first decreasing and then increasing,indicating a correlation between LsCAT1∕2 and resistance to powdery mildew in bottle gourd.Our study provides a theoretical basis for the study and application of CAT genes in the breeding and mechanism of disease resistance in bottle gourd.
