摘 要:目的:研究八角枫科八角枫属植物稀花八角枫Alangium chinense subsp.pauciflorum的化学成分及其体外抗炎活性。方法:采用多种色谱技术对稀花八角枫70%乙醇提取物进行分离纯化,并根据化合物的波谱数据进行结构鉴定。采用脂多糖(LPS)诱导的小鼠巨噬细胞RAW 264.7体外炎症模型测定化合物对一氧化氮(NO)释放的抑制作用。结果:从稀花八角枫的醇提物中分离得到30个化合物,分别鉴定为:八角枫碱(1)、尿嘧啶(2)、尿嘧啶苷(3)、5-羟基-2-羟基甲基吡啶(4)、β-腺苷(5)、5-(hydroxymethyl)-1H-pyrrole-2-carbaldehyde(6)、N-trans-cinnamoyltyramine(7)、6-hydroxy-3,4-dihydro-1-oxo-β-carboline(8)、8-羟基喹啉(9)、(-)-(2R)-1-O-β-D-glucopyranosyl-2-{2,6-dimethoxy-4-[1-(E)-propen-3-ol]phenoxyl}propane-3-ol(10)、愈创木基甘油-9-O-β-D-吡喃葡萄糖苷(11)、4-O-(glycer-2-yl)-dihydroconiferyl alcohol-1′-O-β-D-mannopyranoside(12)、3,5-dimethoxy-4-O-β-D-glucopyranosylphenylpropane-7,9-diol(13)、4-(2,3-dihydroxypropyl)-2,6-dimethoxyphenyl-β-D-glucopyranoside(14)、7-(3-ethoxy-5-methoxyphenyl)propane-7,8,9-triol(15)、4-(α-L-鼠李吡喃糖基氧基)苯甲醛(16)、对羟基苯甲醛(17)、丁香醛(18)、对羟基苯甲酸乙酯(19)、(R)-6-hydroxy-4,7-dimethyl-3,4-dihydronaphthalen-1(2H)-one(20)、对羟基苯乙酮(21)、二氢松柏醇(22)、邻苯二酚(23)、dibothrioclininsⅡ(24)、水杨苷(25)、2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucopyranoside(26)、2-(ethoxymethyl)phenol-1-O-β-D-glucopyranoside(27)、3′-O-methyl-3,4-di-O-methylenedioxyellagic acid(28)、岩白菜素(29)、3′-O-methyl-3,4-methylenedioxyellagic acid-4′-O-β-D-glucopyranoside(30)。结论:其中,化合物3~10、12~22、24、26、27、29为首次从八角枫属植物中分离得到,化合物1、2、11、23、25、28、30为首次从稀花八角枫中分离得到。化合物4、5、7、11~13具有一定的抗炎活性。Objective:To study the chemical constituents of Alangium chinense subsp.pauciflorum and anti-inflammatory activity in vitro.Methods:The 70%ethanol extract of Alangium chinense subsp.pauciflorum was isolated and purified by various chromatographic techniques,the structures were identified on the basis of spectral data.The inhibitory effect of the compounds on nitric oxide(NO)release was determined by lipopolysaccharide(LPS)-induced inflammation model of mouse macrophage RAW 264.7 in vitro.Results:Thrity compounds were isolated from the ethanol extract of Alangium chinense subsp.pauciflorum and identified as anabasine(1),uracil(2),uridine(3),5-hydroxyl-2-hydroxymethylpyridine(4),β-adenosine(5),5-(hydroxymethyl)-1H-pyrrole-2-carbaldehyde(6),N-trans-cinnamoyltyramine(7),6-hydroxy-3,4-dihydro-1-oxo-β-carboline(8),8-hydroxyquinoline(9),(-)-(2R)-1-O-β-D-glucopyranosyl-2-{2,6-dimethoxy-4-[1-(E)-propen-3-ol]phenoxyl}propane-3-ol(10),uaiacylglycerol-9-O-β-D-glucopyranoside(11),4-O-(glycer-2-yl)-dihydroconiferyl alcohol-1′-O-β-D-mannopyranoside(12),3,5-dimethoxy-4-O-β-D-glucopyranosylphenylpropane-7,9-diol(13),4-(2,3-dihydroxypropyl)-2,6-dimethoxyphenyl-β-D-glucopyranoside(14),7-(3-ethoxy-5-methoxyphenyl)propane-7,8,9-triol(15),4-(α-L-rhamnopyranosyloxy)benzaldehyde(16),p-hydroxy-benzaldehyde(17),syringaldehyde(18),p-hydroxybenzoic acid ethyl ester(19),(R)-6-hydroxy-4,7-dimethyl-3,4-dihydronaphthalen-1(2H)-one(20),4-hydroxyacetophenone(21),dihydroconiferylalcohol(22),catechol(23),dibothrioclininsⅡ(24),salicin(25),2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucopyranoside(26),2-(ethoxymethyl)phenol-1-O-β-D-glucopyranoside(27),3′-O-methyl-3,4-di-O-methylenedioxyellagic acid(28),bergenin(29),3′-Omethyl-3,4-methylenedioxyellagic acid-4′-O-β-D-glucopyranoside(30),respectively.Conclusion:Among them,compounds 3 to 10,12 to 22,24,26,27,29 are isolated from Alangium for the first time,compounds 1,2,11,23,25,28,30 are isolated from Alangium chinense subsp.pauciflorum for the first time,compounds 4,5,7,11 to 13 show
