2株小熊猫源色杆菌的鉴定及其耐药性和致病性分析

摘  要:目的对分离自小熊猫内脏的2株菌进行分类鉴定及耐药性与致病性分析。方法从形态学观察、生化特性检测、通过16S rRNA及全基因组序列分析对病原菌进行鉴定和分类,并进行药敏试验、致病性试验、耐药基因预测。结果从小熊猫体内分离的2株菌分别命名为CV08与CV20,经鉴定CV20为紫色色杆菌,CV08是色杆菌新种;药敏试验结果显示2株菌对大多数的β-内酰胺类抗生素不敏感,氨基糖苷类与氟喹诺酮类抗生素高敏;全基因组序列分析结果显示,2株菌均携带多个耐药基因;小鼠感染试验结果显示2株菌具有高致病性,CV20毒性强于CV08。结论分离自小熊猫的2株色杆菌有很强的致病性,具有多重耐药性,本研究为小熊猫色杆菌病的研究与防控奠定基础。Chromobacterium are gram-negative,strictly aerobic or facultatively anaerobic bacteria widely found in water and soil in subtropical and tropical areas.These bacteria occasionally cause human and animal infections with very high mortality rates in the absence of appropriate antibiotic treatment.Here,two strains,CV08 and CV20,were isolated from the internal organs of red pandas.Bacteria were identified and classified through morphological observation,biochemical characteristics,PCR identification,and sequencing analysis.The pathogenic bacteria were identified and analyzed with drug sensitivity tests,pathogenicity tests,and antibiotic resistance gene forecasting.According to 16S rRNA sequence analysis,the similarity between CV20 and known Chromobacterium violaceum strains exceeded 98.65%,and the similarity between CV08 and Chromobacterium sp.ATCC 53434 was 99.19%.According to genome-wide average nucleotide identity analysis,ANI values between CV20 and seven known strains of Chromobacterium violaceum all exceeded 98%,and the ANI value between CV08 and Chromobacterium sp.ATCC 53434 was 92.65%.From the above results,CV08 was identified as a new species of Chromobacterium,and CV20 was identified as Chromobacterium violaceum.Drug sensitivity tests showed that the two strains were highly multi-drug resistant,not sensitive to mostβ-lactam antibiotics,and highly sensitive to aminoglycoside and fluoroquinolone antibiotics.Whole genome sequence analysis indicated that the two strains carried multiple drug resistance genes.Mouse infection tests showed that the two strains of bacteria had high pathogenicity,and the virulence of CV20 was higher than that of CV08.This study lays a foundation for research and control efforts for red panda chromobacteriosis.
