莱州湾南岸第四系沉积物微生物群落垂向变化特征

摘  要:为了探究海水入侵背景下沉积物剖面内微生物群落结构及多样性的垂向变化规律,采集山东省潍坊市莱州湾南岸一眼钻孔深度为200 m的沉积物样品,基于高通量测序方法分析沉积物微生物的16S rRNA基因。结果表明:沉积物全盐量随着深度的增加而逐渐减小,酸碱度随深度的增加而逐渐增大,微生物丰度和α多样性随着深度的增加而降低,变形菌Proteobacteria广泛存在于沉积物中,厚壁菌Firmicutes和拟杆菌Bacteroidete随深度的增加而增加,放线菌Actinobacteria、绿湾菌Chloroflexi、酸杆菌Acidobacteria等随深度的增加而减少;沉积物微生物群落结构的变化特征受多种环境要素共同影响,对微生物群落结构影响最大的是深度和酸碱度,其次是全盐量和总氮。To explore the vertical changes of microbial community structure and diversity in sediment profiles under the background of transgression,16S rRNA gene analysis of sediment microbe was conducted based on high-throughput sequencing method in sediments from a 200 m deep well in the South Bank of Laizhou Bay in Weifang City,Shandong Province.The results show that the total salt content of sediments gradually decreases with increasing depth,and pH increases with depth.The microbial abundance andαdiversity decrease with the increase of depth.Proteobacteria exists widely in the sediment,Firmicutes and Bacteroidete increase with the increase of depth.Actinobacteria,Chloroflexi,and Acidobacteria decrease with the increase of depth.The characteristics of microbial community structure in sediments are affected by various environmental factors,and depth and pH are the most important factors,followed by total salt content and total nitrogen.
