摘 要:为了探究β1,4-半乳糖基转移酶1(β4GalT1)在气道平滑肌细胞中的功能,通过高通量基因表达数据库(GEO)分析β4GalT1在气道平滑肌细胞炎症反应存在功能,采用脂多糖刺激气道平滑肌细胞后检测白介素1β及β4GalT1的表达;采用慢病毒包装构建过表达β4GalT1的稳转细胞,并利用实时荧光定量聚合酶链式反应检测稳转细胞中炎症相关细胞因子表达随时间的变化,同时通过细胞增殖实验、细胞周期和划痕实验检测稳转细胞的生物学行为变化,研究过表达β4GalT1的细胞分泌液在调节细胞因子表达过程中的作用。结果表明:脂多糖能引起白介素1β及β4GalT1的表达量增加;随着培养时间的延长,过表达β4GalT1的细胞中促炎因子表达量减小,抑炎因子表达量增加,同时过表达β4GalT1可改变气道平滑肌细胞的增殖、迁移行为,进而起到炎症修复功能;过表达β4GalT1的细胞可通过分泌液调节细胞因子的表达,表明β4GalT1能快速响应对气道平滑肌细胞的炎症刺激,并通过分泌物质调节气道平滑肌细胞的细胞行为及细胞因子变化。To explore the function ofβ1,4-galactosyltransferase 1(β4GalT1)in airway smooth muscle cells,the function ofβ4GalT1 in the inflammatory response of airway smooth muscle cells was analyzed through Gene Expression Omnibus(GEO)database,and the expression of interleukin 1βandβ4GalT1 after stimulating airway smooth muscle cells with lipopolysaccharide was detected.Lentivirus packaging was used to construct stable cells overexpressingβ4GalT1,and real-time fluorescence quantitative polymerase chain reaction was used to detect the changes in the expression of inflammatory cytokines in stable cells over time,and cell proliferation assay,cell cycle assay and scratch assay were used to detect the biological behavior changes of stable cells,and the role of cell secretions overexpressingβ4GalT1 in regulating cytokine expression was researched.The results show that lipopolysaccharide increases the expression of interleukin 1βandβ4GalT1.With the increase of culture time,the expression of pro-inflammatory factor decreases and the expression of anti-inflammatory factor increases in the cells overexpressingβ4GalT1.Meanwhile,the overexpression ofβ4GalT1 can change the proliferation and migration behavior of airway smooth muscle cells,and play a role in inflammation repair.Cells of overexpressβ4GalT1 can regulate cytokine expression through secretory fluid,which suggest thatβ4GalT1 can rapidly respond to inflammatory stimulation of airway smooth muscle cells,and regulate the cell behavior and cytokine changes of airway smooth muscle cells by secreting substances.
