摘  要:目的研究虎黄烧伤搽剂通过调控Wnt/β-catenin信号通路影响皮肤创伤后小鼠miR-146a表达及相关炎性因子水平。方法选择150只雄性小鼠,随机分组,即对照组(30只)、模型组(30只)、用药组(低剂量组、中剂量组、高剂量组各30只)。在构建小鼠创伤模型后,模型组不用药,低剂量组、中剂量组、高剂量组分别给予10 mg/kg、20 mg/kg、30 mg/kg的虎黄烧伤搽剂,在建模后的第1、3、7、11、14、17天进行观察实验。采用蛋白免疫印迹(Western blot,WB)法检测β-连环蛋白(β-catenin)、糖原合成酶激酶-3β(Glycogen synthase kinase-3,GSK-3β)蛋白表达情况。实时荧光定量PCR(Quantitative real-time-polymerase chain reaction,qRT-PCR)检测miR-146a表达水平。采用酶联免疫吸附实验(Enzyme linked immunosorbent assay,ELISA)法检测白细胞介素4(Interleukin-4,IL-4)、白细胞介素6(Interleukin-6,IL-6)、转化生长因子-β(Transforming growth factor-β,TGF-β)、肿瘤坏死因子-α(Tumor necrosis factor-α,TNF-α)、干扰素-γ(Interferon-γ,IFN-γ)炎性因子水平。结果与对照组相比,模型组β-catenin、GSK-3β蛋白表达水平均升高,miR-146a表达水平降低,血清IL-4、IL-6、TNF-α、TGF-β和IFN-γ炎症因子水平上升,且miR-146a和血清炎症因子的变化程度较大,而用药组β-catenin、GSK-3β蛋白表达水平均则下降,miR-146a表达水平和5种血清炎症因子水平虽仍为下降和上升,但变化幅度远小于模型组(P<0.05),并且不同用药组(低剂量组、中剂量组、高剂量组)间以上各指标差异均存在统计学意义(P<0.05)。结论虎黄烧伤搽剂能通过抑制Wnt/β-catenin信号通路的表达,抑制皮肤创伤后小鼠miR-146a表达水平的降低和血清炎性因子水平的上升。Objective To study the effect of Huhuang Burn Liniment(虎黄烧伤搽剂)on the expressions of miR-146a and related inflammatory factors in mice after skin trauma by regulating Wnt/β-catenin signaling pathway.Methods A total of 150 male mice were selected and randomly divided into different groups,namely the control group(30 rats),model group(30 rats),treatment groups[Huhuang Burn Liniment low dose group(30 mice),Huhuang Burn Liniment middle dose group(30 rats),Huhuang Burn Liniment high dose group(30 mice)].After the trauma model of mice was constructed,the model group was not treated,and the low dose group,middle dose group and high dose group were given 10 mg/kg,20 mg/kg and 30 mg/kg Huhuang Burn Liniment respectively,and the observation experiments were conducted on the 1 st,3 rd,7th,11 th,14th,and 17th day after modeling.The protein expressions ofβ-catenin and glycogen synthase kinase-3β(GSK-3β)were detected by Western blot.Real-time fluorescence quantitative PCR(qRT-PCR)was used to detect the expressions of miR-146a.Enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of interleukin 4(IL-4),interleukin 6(IL-6),transforming growth factor-β(TGF-β),tumor necrosis factor-α(TNF-α)and interferon-γ(IFN-γ).Results Compared with those of the control group,the expressions ofβ-catenin and GSK-3βprotein in the control group were increased,the expression of miR-146a was decreased,and the levels of serum IL-4,IL-6,TNF-α,TGF-βand IFN-γinflammatory factors were increased,and the changes of miR-146a and serum inflammatory factors were greater,while the expressions ofβ-catenin and GSK-3βprotein in the Huhuang Burn Liniment treatment groups were decreased.Although the expression of miR-146a was decreased and the levels of five serum inflammatory factors were still increased,the change was far less than that of the model group(P<0.05),and there were significant differences in the above indexes among different drug groups(low dose group,middle dose group and high dose group)(P<0.05).Conclusion
