摘 要:目的通过观察眼针对脑缺血再灌注损伤(Cerebral ischemia-reperfusion injury,CIRI)模型大鼠脑组织自噬相关蛋白表达的影响,探讨眼针与CIRI神经细胞自噬的关系。方法将120只大鼠随机分为正常组、假手术组、造模组3组,采用改良线栓法对造模组大鼠进行模型复制,造模成功的大鼠分为模型组、3-甲基腺嘌呤(3-methyladenine,3-MA)组、眼针组和3-MA+眼针组。最终,正常组、假手术组和模型组,每组16只;3-MA组、眼针组、3-MA组+眼针组,每组17只。采用ZeaLonga评分法进行大鼠神经功能评分,采用透射电镜观察眼针干预前后CIRI自噬小体的形成情况;采用Western blot法及免疫组化方法观察脑组织LC3、ATG1、ATG4的表达水平。结果与模型组相比,眼针组大鼠神经功能缺损评分明显下降;自噬小体数目减少;大鼠脑组织LC3、ATG1、ATG4蛋白表达显著下降,差异具有统计学意义(P<0.05)。结论眼针疗法能够改善脑缺血再灌注损伤大鼠神经功能缺损评分;减少大鼠脑组织自噬小体数量;其机制可能是通过降低大鼠脑组织LC3、ATG1、ATG4蛋白表达来调控神经细胞过度自噬,从而改善大鼠脑缺血再灌注损伤。Objective To investigate the relationship between eye acupuncture and neuronal autophagy of cerebral ischemia-reperfusion injury(CIRI)by observing the effect of eye acupuncture on the expression of autophagy related proteins in brain tissue of rats with CIRI model.Methods A total of 120 rats were randomly divided into three groups:normal group,sham operation group,model group.The rats in the model group were replicated by modified suture method.The rats that successfully underwent modeling were further divided into model group,3-methyladenine(3-MA)group,eye acupuncture group,and 3-MA+eye acupuncture group.Finally,there were 16 rats in each of normal group,sham operation group and model group;while 3-MA group,eye acupuncture group,and 3-MA+eye acupuncture group each had 17 rats.The ZeaLonga scoring method was used to assess the neurological function of rats,and transmission electron microscopy was used to observe the formation of CIRI autophagosomes before and after eye acupuncture intervention.Western blot and immunohistochemical methods were used to observe the expression levels of LC3,ATG1 and ATG4 in brain tissue.Results Compared with those of the model group,the scores of neurological deficits in the eye acupuncture group were significantly reduced.The number of autophagosomes decreased.The expression of LC3,ATG1 and ATG4 proteins in rat brain tissue significantly decreased with statistical significance(P<0.05).Conclusion Eye acupuncture therapy can improve the neurological deficit score in rats with cerebral ischemia-reperfusion injury.The number of autophagosomes in rat brain tissue was reduced.The mechanism may be to regulate excessive autophagy of neural cells by reducing the expressions of LC3,ATG1 and ATG4 proteins in rat brain tissue,thereby improving CIRI in rats.
